摘 要:目的评价不同炮制方法对黄药子抗胃癌作用及其肝毒性的影响。方法80只小鼠随机分为空白对照(Ct)组、黄药子生品(RD)组、清炒黄药子(FD)组、酒炙黄药子(WD)组、当归煎汤炙黄药子(2∶1)(ASR2∶1DB)组、黄芪煎汤炙黄药子(1∶2)(AR1∶2DB)组、黄芪煎汤炙黄药子(1∶1)(AR1∶1DB)组、黄芪煎汤炙黄药子(2∶1)(AR2∶1DB)组,每组各10只,各组均以1.7g·kg^(-1)剂量灌胃给予对应炮制药物,连续14d,Ct组给予等量生理盐水。观察和比较各组肝脏病理形态学变化。体外培养人胃癌细胞MKN-45,以各组含药血清处理,CCK-8法检测细胞增殖情况。结果病理形态学变化表明各炮制组均造成了不同程度的肝脏损伤,各给药组肝脏病变损害程度按由轻到重依次为AR2∶1DB、ASR2∶1DB、AR1∶1DB、AR1∶2DB、FD、WD、RD组,以RD组损害最为严重;CCK-8实验结果表明AR1∶1DB、AR2∶1DB、ASR2∶1DB组可不同程度抑制人胃癌细胞MKN-45增殖(P<0.05),以AR2∶1DB组作用最强(P<0.001)。FD、WD、AR1∶2DB组间人胃癌细胞MKN-45增殖抑制作用无统计学差异;结论黄芪煎汤炙黄药子炮制法可增强黄药子体外抗胃癌活性并减少其对肝脏的损害,药效有剂量依赖性,随着黄芪在药对中所占的比重增大而增强,以2∶1组效果最佳,其机制可能与增强抗氧化及清除自由基作用有关。Objective To evaluate the influence of different processing methods on the anti-gastric cancer effect and hepatotoxicity of Dioscorea bulbifera L.Methods 80 mice were randomly divided into blank control(Ct),raw Dioscorea bulbifera L.(RD),fried Dioscorea bulbifera L.(FD),wine-fried Dioscorea bulbifera L.(WD),Angelicae sinensis radix liquid-fried Dioscorea bulbifera L.(2∶1)(ASR2∶1DB),Astragali radix liquid-fried Dioscorea bulbifera L.(1∶2)(AR1∶2DB),Astragali radix liquid-fried Dioscorea bulbifera L.(1∶1)(AR1∶1DB)and Astragali radix liquid-fried Dioscorea bulbifera L.(2∶1)(AR2∶1DB),10 in each group.Mice in 8 groups were administered with corresponding processed drugs given in gavage respectively at a dose of 1.7g·kg^(-1) for consecutive 14d and equal amounts of normal saline were administered in the Ct group.Liver pathomorphological changes were observed and compared.MKN-45 human gastric cancer cells were cultured in vitro.Each group was treated with serum containing drugs and cell proliferation was detected by the CCK-8 assay.Results Pathomorphological changes showed that each processed group caused different degrees of liver damage,the degree of liver lesions in each administration group was AR2∶1DB,ASR2∶1DB,AR1∶1DB,AR1∶2DB,FD,WD,and RD from light to heavy with the most severe damage in the RD group;CCK-8 results showed that the AR1∶1DB,AR2∶1DB and ASR2∶1DB groups can inhibit MKN-45 human gastric cancer cells proliferation in different degrees(P<0.05),the strongest effect among these three groups was observed in the AR2∶1DB group(P<0.001).There was no statistical difference in the inhibition of MKN-45 proliferation by FD,WD and AR1∶2DB.Conclusion The processing of astragali radix liquid-fried Dioscorea bulbifera L.can enhance the anti-gastric cancer effect of Dioscorea bulbifera L.and reduce its damage to the liver.Its efficacy is dose-dependent with the best effect of the 2∶1 group.The mechanism may be related to the enhancement of antioxidant and free radical clearance.
