摘 要:目的:观察电针心包经穴对中动脉阻塞(middle cerebral artery occlusion, MCAO)模型大鼠缺血脑组织D-丝氨酸(D-serine, D-Ser)含量和N-甲基-D-天冬氨酸(N-methyl-D-aspartic Acid, NMDA)受体亚单位NR1蛋白表达的影响。方法:将120只SD大鼠随机分为电针组和模型组,每组再分1 d、3 d、7 d、14 d、21 d五个时相点,每时相点12只,采用线栓法制备MCAO模型大鼠。电针组给予电针心包经穴,模型组仅捆绑处理,每次30 min,每天1次。采用走横木实验对处理前后大鼠神经功能缺损情况评分,高效液相色谱(high performance liquid chromatography, HPLC)检测大鼠缺血脑组织D-Ser含量,Western Blot法检测大鼠缺血脑组织NR1蛋白表达。结果:处理后同一时相点与模型组比较,电针组3 d、7 d、14 d、21 d走横木评分明显增高(P<0.05),3 d、14 d、21 d组脑组织D-Ser含量明显降低(P<0.05),五个时相点脑组织NR1蛋白表达均明显降低(P<0.05)。结论:电针心包经穴可能通过降低MCAO模型大鼠缺血脑组织D-Ser含量、抑制NR1过度表达而发挥神经保护作用。Objective: To observe the effect of electroacupuncture at pericardial Meridian on the content of D-serine(D-Ser) and the expression of NR1 protein of N-methyl-D-aspartic acid(NMDA) receptor subunit in ischemic brain tissue of rats with middle cerebral artery occlusion(MCAO).Methods: 120 SD rats were randomly divided into electroacupuncture group and model group.Each group was divided into five phase points: 1,3,7,14 and 21 days, with 12 rats at each phase point.MCAO model rats were prepared by thread bolt method.The electroacupuncture group was given electroacupuncture at pericardial meridian points, and the model group was only bound for 30 minutes each time, once a day.The level of D-Ser in ischemic brain tissue was detected by high performance liquid chromatography(HPLC),and the expression of NR1 protein in ischemic brain tissue was detected by Western blot.Results: After treatment, compared with the model group at the same time point, the walking cross beam score of electroacupuncture group was significantly higher at 3,7,14 and 21 days(P<0.05),the content of D-Ser in brain tissue was significantly lower at 3,14 and 21 days(P<0.05),and the expression of NR1 protein in brain tissue was significantly lower at five time points(P<0.05).Conclusion: Electroacupuncture at pericardial meridian points may play a neuroprotective role by reducing the content of D-Ser in ischemic brain tissue and inhibiting the overexpression of NR1 in MCAO model rats.
