摘 要:目的:探讨miRNA-27a rs895819基因多态性与妊娠期糖尿病的相关性。方法:选取2020年1月~8月在广东省佛山市妇幼保健院就诊并诊断为GDM的孕妇和正常孕妇各200例,测定两组孕妇血糖、胆固醇(CHOL)、甘油三脂(TG)、C反应蛋白(CRP)和胰岛素水平,计算胰岛素抵抗指数(HOMA-IR)和胰岛β细胞功能指数(HOMA-β),并进行统计分析;采用聚合酶链反应-连接酶检测反应(PCR-LDR)法检测两组孕妇rs895819位点的基因型,比较该位点等位基因和基因型的分布差异;分析两组孕妇不同基因型临床生化指标的差异。结果:两组孕妇的年龄、空腹血糖(FBG)、1小时血糖(1h-PG)、2小时血糖(2h-PG)、CHOL、TG、TG、HOMA-IR和HOMA-β比较差异有统计学意义(P<0.05);rs895819位点CC、CT和TT基因型分布频率在GDM组分别是4.5%、39%和56.5%,对照组分别是5.5%、41.2%和53%,两组基因型分布差异无统计学意义(P>0.05);与CC基因型相比,TT基因型的2h-PG、HOMA-IR和HOMA-β均升高。结论:未发现miRNA-27a rs895819基因多态性与广东佛山地区妊娠期糖尿病的相关性。Objective:To explore the association between miRNA-27a rs895819 gene polymorphism and gestational diabetes mellitus.Methods:A total number of 400 pregnant women in our hospital from January to August 2020 were enrolled.200 pregnant women with GDM were selected as GDM group and 200 normal pregnant women as the control group.The level of blood glucose、CHOL、TG、CRP and insulin were analyzed.Genotyping was detected by polymerase chain reaction-ligase detection(PCR-LDR).The relationship between genotype distributions of rs895819 and biochemical data were analyzed.Results:There were significant differences in age,FBG,1h-PG,2h-PG,CHOL,TG,TG,HOMA-IR and HOMA-β between the two groups(P<0.05),but there were no significant difference in CRP and insulin(P>0.05).The CC,CT and TT genotype frequencies of rs895819 in GDM group were 4.5%、39%and 56.5%,which in the control group were 5.5%、41.2%and 53%.There was no significant correlation of genotype frequencies between two groups(P>0.05).In addition,the levels of 2h-PG、HOMA-IR and HOMA-β in pregnant with rs895819 TT genotype were significantly higher than those with CC genotype.Conclusion:MiRNA-27a rs895819 gene polymorphism has no correlation with gestational diabetes mellitus in Guangdong.
