产长春胺类生物碱内生真菌CH1原生质体灭活条件的研究

摘  要:长春胺类生物碱是一类广泛存在于夹竹桃科植物中的吲哚生物碱,研究发现该类生物碱对老年痴呆、脑动脉硬化、脑梗死、脑卒中等多种疾病具有很好的治疗作用,尤其长春西汀还可作为膳食补充剂,具有广阔的发展前景。本实验以CH1为亲本菌株,研究了热灭活和紫外灭活两种不同的灭活方法对内生真菌CH1原生质体再生的影响,确定CH1原生质体的最佳灭活条件为:在距30 W紫外灯20 cm处照射10 min,CH1亲本原生质体灭活率可达100%,在60℃水浴处理25 min时,内生真菌CH1原生质体灭活率达100%,这为提高内生真菌CH1原生质体融合后融合子的筛选效果奠定一定基础。Vincamine indole alkaloids, the most common nitrogen-containing secondary metabolites, are widely present in plants of the Apocynaceae family. It has been found that this type of alkaloids had good effects on various diseases such as Alzheimer’s disease, cerebral arteriosclerosis, cerebral infarction and stroke. Vinpocetine can also be used as a dietary supplement, which has a broad development prospect. In this paper, the effects of heat inactivation and UV inactivation on protoplast regeneration of endophytic fungus CH1 were studied. The optimal conditions for asymmetrically inactivated protoplasts of CH1 were as follows: CH1 was inactivated by heat for 25 min and UV for 10 min respectively. The result laid a certain foundation for improving the screening effect of fusion of endophytic fungi CH1 protoplasts.
