摘 要:目的:探讨穿心莲内酯对胰腺癌细胞顺铂敏感性的作用和机制。方法:将人胰腺癌细胞SW1990分成对照组、穿心莲内酯组(穿心莲内酯处理)、顺铂组(顺铂处理)、穿心莲内酯+顺铂组(穿心莲内酯、顺铂处理)、穿心莲内酯+顺铂+转化生长因子-β_(1)(TGF-β_(1))组(穿心莲内酯、顺铂、TGF-β_(1)处理)、TGF-β_(1)组(TGF-β_(1)处理),Western Blotting检测TGF-β_(1)、p-Smad2/Smad2、p-Smad3/Smad3蛋白表达,MTT检测增殖,流式细胞术检测凋亡,Transwell小室检测侵袭和迁移。结果:与对照组比较,穿心莲内酯组胰腺癌细胞TGF-β_(1)、p-Smad2/Smad2、p-Smad3/Smad3蛋白表达量减少(P<0.05),TGF-β_(1)组胰腺癌细胞p-Smad2/Smad2、p-Smad3/Smad3蛋白表达量增多(P<0.05)。与对照组比较,穿心莲内酯组、顺铂组、穿心莲内酯+顺铂组胰腺癌细胞增殖活性减少,凋亡率增多,细胞侵袭和迁移数目降低(P<0.05)。与穿心莲内酯组、顺铂组比较,穿心莲内酯+顺铂组胰腺癌细胞增殖活性减少,凋亡率增多,细胞侵袭和迁移数目降低(P<0.05)。与穿心莲内酯+顺铂组比较,穿心莲内酯+顺铂+TGF-β_(1)组胰腺癌细胞增殖活性增多,凋亡率减少,细胞侵袭和迁移数目增多(P<0.05)。结论:穿心莲内酯通过抑制TGF-β_(1)/Smad2/3信号通路提高胰腺癌细胞对顺铂敏感性。Objective:To investigate the effect and mechanism of andrographolide on the sensitivity of pancreatic cancer cells to cisplatin.Methods:Human pancreatic cancer cells SW1990 were divided into a control group,an andrographolide group(andrographolide treatment),cisplatin group(cisplatin treatment),an andrographolide+cisplatin group(andrographolide,cisplatin treatment),andrographolide+cisplatin+transforming growth factorβ_(1)(TGF-β_(1))group(andrographolide,cisplatin,TGF-β_(1) treatment),a TGF-β_(1) group(TGF-β_(1) treatment),Western blot detection of TGF-β_(1),p-Smad2/Smad2,p-Smad3/Smad3 protein expression,MTT detection of proliferation,flow cytometry detection of apoptosis,Transwell chamber detection of invasion and migration.Results:Compared with the control group,TGF-β_(1),p-Smad2/Smad2,p-Smad3/Smad3 protein expression in pancreatic cancer cells in the andrographolide group decreased(P<0.05).Compared with the control group,the expression of p-Smad2/Smad2,p-Smad3/Smad3 protein in pancreatic cancer cells in the TGF-β_(1) group increased(P<0.05).Compared with the control group,pancreatic cancer cell proliferation activity decreased,apoptosis rate increased,the number of cell invasion and migration decreased in andrographolide group,cisplatin group,and andrographolide+cisplatin group(P<0.05).Compared with the andrographolide group and the cisplatin group,the pancreatic cancer cell proliferation activity in the andrographolide+cisplatin group decreased,and the apoptotic rate increased,the number of cell invasion and migration decreased(P<0.05).Compared with andrographolide+cisplatin group,pancreatic cancer cell proliferation activity increased,apoptosis rate decreased,the number of cell invasion and migration increased in andrographolide+cisplatin+TGF-β_(1) group(P<0.05).Conclusion:Andrographolide can increase the sensitivity of pancreatic cancer cells to cisplatin by inhibiting the TGF-β_(1)/Smad2/3 signaling pathway.
