摘 要:目的探讨花椒根总生物碱的抗肿瘤活性。方法实验分为6组,各10只,分别灌胃1000,800,640,512,410,328 g/kg花椒根总生物碱1次(每10 g体质量灌胃0.2 mL),计算半数致死浓度(LD_(50))。将60只KM小鼠随机分为正常对照组(A组,等体积生理盐水灌胃)、模型组(B组,等体积生理盐水灌胃)、阳性药物组(C组,氟尿嘧啶20 mg/kg腹腔注射)及花椒根总生物碱高、中、低剂量组(D_(1)组、D_(2)组、D_(3)组,生药100,80,40 g/kg灌胃),各10只,右上肢腋窝皮下注射单细胞悬液0.2 mL以复制H22荷瘤小鼠模型,建模成功后分别给予相应药物或生理盐水,每天1次,连续10 d。采用酶联免疫吸附(ELISA)法检测小鼠血清中白细胞介素2(IL-2)及肿瘤坏死因子-α(TNF-α)水平;剥取瘤块,称定质量,计算抑瘤率;苏木素-伊红(HE)染色,观察瘤体组织病理形态学;Tunel染色,观察H22肿瘤细胞凋亡情况。结果与B组比较,C组和D_(1)组小鼠瘤体质量显著减轻(P<0.05);D_(1)组小鼠血清中IL-2和C组、D_(1)组、D_(2)组小鼠血清中TNF-α水平均显著升高(P<0.05或P<0.01);C组、D_(1)组、D_(2)组细胞凋亡率显著升高(P<0.01);C组、D_(1)组、D_(2)组、D_(3)组小鼠的抑瘤率分别为53.26%,36.85%,10.29%,2.18%。B组肿瘤细胞数目多;排列紧密;细胞核深染;C组和D_(1)组肿瘤细胞胞核固缩、胞质疏松,可见凋亡、坏死的肿瘤细胞。结论花椒根总生物碱具有体内抗肿瘤作用,其机制可能与提高机体细胞免疫功能及促进肿瘤细胞凋亡有关。Objective To investigate the anti-tumor activity of total alkaloids from Zanthoxyli Radix.Methods Sixty SPF KM mice were randomly divided into six groups,with ten mice in each group.Mice in each group were gavaged with 1000,800,640,512,410 and 328 g/kg of total alkaloids from Zanthoxyli Radix once(0.2 mL per 10 g of body weight).The median lethal concentration(LD_(50))was calculated.Sixty KM mice were randomly divided into the normal control group(group A,intragastric administration of equal volume of normal saline),model group(group B,intragastric administration of equal volume of normal saline),positive drug group(group C,intraperitoneal injection of 20 mg/kg of fluorouracil),and high-,medium-and low-dose groups of total alkaloids from Zanthoxyli Radix(groups D_(1),D_(2) and D_(3),intragastric administration of 40,80,100 g/kg of crude drug),with 10 mice in each group.H22 tumor-bearing mouse model was established by subcutaneous injection of 0.2 mL of single-cell suspension into the armpit of the right upper limb of mice.After successful modeling,the mice were given corresponding drugs or normal saline once a day for 10 d.The levels of interleukin-2(IL-2)and tumor necrosis factor-α(TNF-α)in serum were measured by the enzyme-linked immunosorbent assay(ELISA).The tumor was stripped and weighed,and the tumor inhibition rate was calculated.Hematoxylin-eosin(HE)staining was used to observe the histopathology of tumor tissue.Tunel staining was used to observe the apoptosis of H22 tumor cells.Results Compared with those in the group B,the weight of tumor in the groups C and D_(1) decreased significantly(P<0.05),the levels of serum IL-2 in the group D_(1) and serum TNF-αin the groups D_(1) and D_(2) increased significantly(P<0.05 or P<0.01),the apoptosis rate in groups C,D_(1) and D_(2) increased significantly(P<0.01),the tumor inhibition rates of mice in groups C,D_(1),D_(2) and D_(3) were 53.26%,36.85%,10.29%and 2.18%,respectively.The tumor cells in group B were numerous and closely arranged,and the nuclei were dee
