摘  要:柯萨奇病毒A组16型(Coxsackievirus A16,CV-A16)是导致手足口病的病原体之一,CV-A16感染后宿主的外周血细胞的细胞因子表达变化的报道较少,也未见有关血清中细胞因子含量在感染前后的表达变化。为了分析CV-A16感染的不同严重程度的沙鼠细胞因子和趋化因子的动态变化。通过腹腔注射CV-A16构建CV-A16感染发病模型,应用流式细胞术、实时荧光定量RT-PCR和ELISA法检测病毒载量、血清炎症因子和趋化因子水平动态变化。CV-A16感染后重症沙鼠的临床症状,包括体重下降程度、临床症状分值以及死亡率均显著高于轻症沙鼠,并且重症沙鼠外周血白细胞比例和升高趋势总体上高于轻症感染沙鼠;重症感染沙鼠全血中细胞因子IL-2、IL-12、IFN-γ和TNF-α的mRNA转录水平上调,IL-4、IL-10、IL-8和IP-10的mRNA转录水平下调;并且CV-A16感染后重症沙鼠血清中的TNF-α和IFN-γ含量显著上升,IL-8和IP-10的含量显著下降。结论为重症沙鼠全血和血清中存在多种趋化和炎症因子上调,可能与Th1炎症失调有关。Coxsackievirus A16(CV-A16)is one of the pathogens of hand,foot and mouth disease,few studies have focused on the change of cytokine expression in peripheral blood cells of the host after CV-A16 infection,and if there is no change in cytokine expression in serum before and after infection.In order to analyze the dynamic changes of expression of cytokines and chemokines in gerbils infected with CV-A16 of different severity.The pathogenesis of CV-A16 infection was constructed by intraperitoneal injection of CV-A16.Flow cytometry,real-time fluorescent reverse transcription-quantitative polymerase chain reaction,and enzyme-linked immunosorbent assay were used to detect the dynamic changes in levels of the viral load,proinflammatory factors,and chemokines.The results show that:the clinical symptoms of gerbils with severe CV-A16 infection(bodyweight loss,clinical symptom scores,and mortality),were significantly higher than those of gerbils with mild CV-A16 infection.The proportion and rising trend of peripheral-blood leukocytes in gerbils with severe infection were,in general,higher than those in gerbils with mild infection.In severely infected gerbils,mRNA transcription of the cytokines interleukin(IL)-2,IL-12,interferon(IFN)-γand tumor necrosis factor(TNF)-αin whole blood was upregulated,and mRNA transcription of IL-4,IL-10,IL-8 and interferon gamma-induced protein(IP)-10 was downregulated.After CV-A16 infection,the serum level of TNF-αand IFN-γin severely infected gerbils increased significantly,and levels of IL-8 and IP-10 decreased significantly.So we can draw a conclusion:various chemotactic and proinflammatory factors show upregulated expression in the whole blood and serum of gerbils with severely infection by CV-A16,which may be related to T-helper type-1 inflammatory disorders.
