摘 要:膜荚黄芪的主要有效成分为黄芪皂苷,为了进一步研究黄芪皂苷生物合成途径及其关键酶的功能,本文利用转录组测序对膜荚黄芪皂苷合成路径中的甲羟戊酸(MVA)途径的关键酶基因进行挖掘,并对氨基酸序列和系统进化树等进行生物信息学分析。结果获得了3-羟基-3-甲基戊二酰辅酶A合成酶(HMGS)和3-羟基-3-甲基戊二酰辅酶A还原酶(HMGR)2个关键酶基因,各6条共12条候选基因序列。本研究结果为膜荚黄芪MVA合成途径的关键酶挖掘以及皂苷合成通路的完全解析提供依据。Saponin in the one of the main effective components in Astragalus membranaceus. In order to further study the biosynthesis pathway of Astragalus saponins and the functions of its key enzymes, the key enzyme genes of mevalonate(MVA) pathway in the biosynthesis pathway of Astragalus membranaceus saponins were excavated based on the genetic information of transcriptome sequencing, and the amino acid sequences and phylogenetic tree were analyzed with the method of bioinformatics. The research results showed that 12 key enzymes genes of 3-hydroxy-3-methylglutaryl CoA synthase(HMGS) and 3-hydroxy-3-methylglutaryl CoA reductase(HMGR) were obtained. The results of this study provide a basis for the excavation of key enzymes in the MVA synthesis pathway of Astragalus membranaceus and the complete analysis of saponin synthesis pathway.
