摘  要:目的基于白细胞介素(IL)-23/辅助性T细胞17(Th17)炎症轴探讨豨莶草对胶原诱导性关节炎(CIA)大鼠模型的治疗作用。方法建立CIA大鼠模型,将建模成功大鼠随机分为模型组、豨莶草组、豨莶草+IL-23组,每组10只;另外10只作为阴性对照组。观察大鼠形态并称量体质量,运用水容积法测定大鼠足趾肿胀度、5级评分法测关节炎指数;苏木精-伊红染色(HE)检测滑膜组织形态学情况;酶联免疫吸附(ELISA)、实时荧光定量PCR(RT-qPCR)检测大鼠腹主动脉血和滑膜组织中IL-23、IL-17、IL-6、IL-22、IL-21含量及mRNA水平;流式细胞术检测腹主动脉血和滑膜组织中Th17细胞百分比。结果与阴性对照组相比,模型组大鼠体质量降低,足趾肿胀度、关节炎指数、腹主动脉血和滑膜组织中IL-23、IL-17、IL-6、IL-22、IL-21含量及mRNA水平、Th17细胞百分比均升高(P<0.05);与模型组相比,豨莶草组大鼠体质量升高,足趾肿胀度、关节炎指数、腹主动脉血和滑膜组织中IL-23、IL-17、IL-6、IL-22、IL-21含量及mRNA水平、Th17细胞百分比均降低(P<0.05);与豨莶草组相比,豨莶草+IL-23组大鼠体质量降低,足趾肿胀度、关节炎指数、腹主动脉血和滑膜组织中IL-23、IL-17、IL-6、IL-22、IL-21含量及mRNA水平、Th17细胞百分比均升高(P<0.05)。结论豨莶草可通过抑制IL-23/Th17炎症轴从而降低炎症水平,实现对CIA大鼠的治疗。Objective To investigate the therapeutic effect of siegesbeckia orientalis on the collagen induced arthritis(CIA)rat model based on interleukin-23(IL-23)/T helper cell 17(Th17)inflammatory axis.Methods The CIA rat model was established.The rats successfully modeled were randomly divided into model group,siegesbeckia orientalis group,siegesbeckia orientalis+IL-23 group,10 in each group,and the other 10 rats in negative control group were treated as control.The morphology and weight of rats were observed,the swelling degree of toes was measured by water volume method,and the arthritis index was measured by 5-grade scoring method;hematoxylin eosin staining(HE)was used to detect the morphology of synovial membrane;the contents and mRNA levels of IL-23,IL-17,IL-6,IL-22,IL-21 in abdominal aortic blood and synovial tissue were detected by enzyme-linked immunosorbent assay(ELISA)and real-time fluorescence quantitative PCR(RT-qPCR);the percentage of Th17 cells in abdominal aortic blood and synovial tissue was detected by flow cytometry.Results Compared with those in the negative control group,the body weight of rats in the model group was lower,the swelling degree of toe,arthritis index,contents and mRNA levels of IL-23,IL-17,IL-6,IL-22,IL-21 in abdominal aortic blood and synovial tissue,and percentage of Th17 cells were higher(P<0.05);compared with those in the model group,the body weight of rats in the siegesbeckia orientalis group was higher,the swelling degree of toe,arthritis index,contents and mRNA levels of IL-23,IL-17,IL-6,IL-22,IL-21 in abdominal aortic blood and synovial tissue,and percentage of Th17 cells were lower(P<0.05);compared with the siegesbeckia orientalis group,the body weight of rats in the siegesbeckia orientalis+IL-23 group was lower,the swelling degree of toe,arthritis index,contents and mRNA levels of IL-23,IL-17,IL-6,IL-22,IL-21 in abdominal aortic blood and synovial tissue,and percentage of Th17 cells were higher(P<0.05).Conclusion Siegesbeckia orientalis can reduce the level of inflammation by
