摘  要:目的:研究nm23核苷二磷酸激酶-1(nm23-H1)与基质金属蛋白酶9(MMP-9)基因在子宫内膜异位症(EMS)患者中的表达方式及意义。方法:选择弋矶山医院30例EMS患者作为研究组,另选取35例正常子宫内膜组织为对照组。通过qRT-PCR技术检测nm23-H1和MMP-9在EMS患者异位内膜及对照组正常内膜组织中mRNA表达;利用Western blot技术检测nm23-H1与MMP-9的蛋白表达。同时分析EMS异位内膜组织中nm23-H1和MMP-9表达量之间的关系。结果:与正常子宫内膜组织相比,异位子宫内膜组织中nm23-H1 mRNA及蛋白水平表达均下调(P<0.001),MMP-9 mRNA及蛋白水平表达均上调(P<0.001),差异有统计学意义;Pearson相关分析显示异位子宫内膜组织中nm23-H1和MMP-9 mRNA表达呈负相关(r=-0.315,P=0.012)。结论:nm23-H1和MMP-9在异位子宫内膜组织中表达异常,可能参与EMS的发病。Objective:To investigate the expression and implications of nm23 nucleoside diphosphate kinase 1(nm23-H1)and matrix metalloproteinases-9(MMP-9)in ectopic endometriosis(EMS).Methods:Ectopic endometrial tissues were obtained in 30 EMS patients(experimental group)from Yijishan Hospital of Wannan Medical College,and normal endometrial tissues were obtained from another 35 women as controls(control group).The mRNA expression of nm23-H1 and MMP-9 in ectopic endometrial tissues and normal endometrial tissues were detected by qRT-PCR.Western blot assay was used to measure the protein expression of nm23-H1 and MMP-9,and the relationship between expression level of nm23-H1 and MMP-9 in ectopic endometrial tissues was analyzed.Results:Compared with normal endometrial tissue,mRNA and protein level of nm23-H1 were down-regulated in ectopic endometrial tissue(P<0.001),whereas mRNA and protein level of MMP-9 were up-regulated(P<0.001).Pearson correlation analysis showed a negative correlation between nm23-H1 and MMP-9 mRNA expression in ectopic endometrium(r=-0.315,P=0.012).Conclusion:nm23-H1 and MMP-9 were abnormally expressed in ectopic endometrium,suggesting that these two genes may be involved in the pathogenesis of endometriosis.
