摘  要:【目的】探究不同种类及质量浓度植物生长调节剂对药用植物米槁愈伤组织诱导、组培苗生根的影响,寻找最适宜米槁愈伤组织诱导和组培苗生根的最佳培养基组合,为米槁大量繁殖提供技术支撑,加速米槁苗木生产,丰富米槁的无性繁殖体系。【方法】以米槁叶片和不定芽为外植体,采用正交试验设计,研究不同种类及质量浓度组合植物生长调节剂对米槁愈伤组织诱导和组培苗生根的影响。【结果】MS培养基适合米槁叶片愈伤组织诱导,不同质量浓度的2,4-D和6-BA均对米槁叶片愈伤组织诱导有显著影响,NAA的质量浓度无显著影响,2,4-D和6-BA是影响愈伤组织诱导的两个主要因素,当2,4-D质量浓度为1.0 mg/L、6-BA质量浓度为2.0 mg/L、NAA质量浓度为0.3 mg/L时,愈伤组织诱导率最高,达到了81.25%。1/2 MS培养基适合米槁不定芽组培苗生根,不同质量浓度的IBA和AC(活性炭)均对米槁不定芽组培苗生根有显著影响,NAA的质量浓度无显著影响,IBA和AC是影响组培苗生根的主要因素,当IBA质量浓度为0.1 mg/L、NAA质量浓度为0.5 mg/L、AC质量浓度为0.2 g/L时,组培苗生根率最高,达到了72.38%,且根系生长状况最好,生根较早,根系繁密,有须根,根长较长。【结论】米槁叶片愈伤组织诱导最佳培养基为MS培养基+2,4-D 1.0 mg/L+6-BA 2.0 mg/L+NAA 0.3 mg/L+琼脂6.5 g/L+蔗糖30 g/L,2,4-D和6-BA比NAA更适合米槁叶片愈伤组织诱导;米槁不定芽组培苗生根最佳培养基为1/2 MS+NAA 0.5 mg/L+IBA 0.1 mg/L+AC 0.2 g/L+琼脂7 g/L+蔗糖30 g/L,IBA和AC比NAA更适合米槁不定芽组培苗生根。【Objective】The work was conducted to explore the effects of different kinds and concentrations of plant growth regulators on the callus induction and tissue culture seedlings rooting of the medicinal plant Cinnamomum migao,in order to screen for the best medium combinations for the callus induction and rooting of C.migao,offering technical support for the mass propagation of C.migao,accelerating its production and enriching the asexual-propagation system.【Method】Taking the leaves and adventitious buds as explants,orthogonal experiments were conducted to study the effects of different kinds and concentrations of plant growth regulators on the callus induction and rooting of tissue cultured C.migao seedlings.【Result】Based on the results,MS medium was most suitable for the callus induction of C.migao leaves,the concentration of 2,4-D and 6-BA had significant effects on the callus induction,while NAA concentration exhibited no significant effect on that.2,4-D and 6-BA were the two main factors affecting the induction.With a concentration of 1.0 mg/L 2,4-D,2.0 mg/L 6-BA and 0.3 mg/L NAA,the callus induction rate showed the highest,reaching 81.25%.Meanwhile,1/2 MS medium was suitable for the rooting of C.migao adventitious buds,the concentration of IBA and AC(activated carbon)had significant effects on the rooting of tissue culture seedlings,while NAA concentration revealed no significant effect on the callus induction of C.migao adventitious buds.IBA and AC were the main factors affecting adventitious buds.When the concentration of IBA,NAA and AC reached 0.1 mg/L,0.5 mg/L and 0.2 g/L,the rooting rate was the highest,reaching 72.38%.In this case,the roots exhibited the best growth,which means dense fibrous long roots with a short rooting period.【Conclusion】The MS medium containing 1.0 mg/L 2,4-D,2.0 mg/L 6-BA,0.3 mg/L NAA,6.5 g/L Agar and 30 g/L Sucrose is the best medium for the callus induction of C.migao leaves.Compared with NAA,2,4-D and 6-BA are more suitable for callus induction.The 1/2 MS medium
