摘 要:研究归芪多糖(GQP)延缓大鼠脑组织衰老的作用机制。将60只SD大鼠随机分为空白对照组(Control)、模型组(Model)、GQP低剂量组(LP)、GQP中剂量组(MP)、GQP高剂量组(HP)、阳性药物白藜芦醇组(Res)。采用皮下注射D-半乳糖(D-gal)建立大鼠衰老模型,实验组和阳性药物组分别给予GQP和Res灌胃。测定脑组织中总超氧化物歧化酶(T-SOD)和谷胱甘肽(GSH)活性、丙二醛(MDA)和ATP质量摩尔浓度;苏木素-伊红(HE)染色观察脑组织病理学变化;检测线粒体膜电位变化以及线粒体膜肿胀程度;免疫组化法检测Bax和Bcl-2蛋白质的表达水平。结果发现:D-gal可致大鼠脑组织中T-SOD、GSH水平和ATP质量摩尔浓度明显降低,MDA质量摩尔浓度显著升高(P<0.05);GQP和Res逆转了上述指标(P<0.05);正常大鼠脑组织未呈现出明显的病理变化,模型大鼠细胞核明显变大、深染,GQP显著减轻了上述病灶;与正常组相比,模型组线粒体膜电位,ATP质量摩尔浓度和Bcl-2蛋白质表达水平显著降低(P<0.01),线粒体膜明显肿胀,Bax表达升高(P<0.01);GQP和Res明显提高了线粒体膜电位、ATP质量摩尔浓度及Bcl-2的表达(P<0.05),同时,减弱了线粒体膜的肿胀,降低了Bax表达(P<0.05)。因此,推测GQP可能通过减轻机体氧化损伤和改变线粒体功能发挥延缓衰老的作用。The mechanism of Guiqi polysaccharides(GQP)in delaying the aging of rat brain tissue was investigated.60 SD rats were randomly divided into the control group(Control),model group(Model),GQP low-dose group(LP),GQP medium-dose group(MP),GQP high-dose group(HP),and positive drug resveratrol group(Res).The aging model of rats was established by subcutaneous injection of D-galactose(D-gal).The experimental group and the positive drug group were orally administered GQP and resveratrol,respectively.The activities of total superoxide dismutase(T-SOD)and glutathione(GSH)were determined and the level of malondialdehyde(MDA)and ATP of ratbrain tissues were measured.The pathological changes of brain tissue were observed by hematoxylin-eosin(HE)staining.The change of mitochondrial membrane potential and the degree of swelling of mitochondrial membrane were detected.In addition,the expression levels of Bax and Bcl-2 were tested by immunohistochemical method.The results showed that D-gal significantly reduced the levels of T-SOD,GSH and ATP in brain tissues of rats,and significantly increased the content of MDA(P<0.05).However,GQP and Res reversed the above indicators(P<0.05).There were insignificant pathological changes of brain tissue in normal rats,but the nuclei in model group were significantly enlarged and deep-stained,and GQP significantly alleviated the above-mentioned lesions.Compared with the control group,the mitochondrial membrane potential,ATP content and Bcl-2 expression were significantly decreased(P<0.01),while the mitochondrial membrane was significantly swollen and the expression of Bax was increased in model group(P<0.01).Meanwhile,GQP and Res increased the mitochondrial membrane potential,ATP content and Bcl-2 expression(P<0.05),but decreased the swelling of mitochondrial membrane and the expression of Bax(P<0.05).Consequently,it was suggested that GQP might play a role in delaying aging by reducing the oxidative damage and changing the mitochondrial function.
