摘 要:目的 研究火炭母体外对人宫颈癌SiHa细胞株增殖抑制的影响。方法 采用CellCounting Kit-8(CCK8)法测定火炭母不同溶剂提取物处理的SiHa细胞增殖活性;采用液质联用色谱法分析火炭母水提液中成分;采用CCK8法测定不同浓度、不同时间柯里拉京处理的SiHa细胞增殖活性,计算IC_(50);采用流式细胞仪检测不同浓度柯里拉京处理48h后的SiHa细胞凋亡率。结果 火炭母水提液体外抑制SiHa细胞增殖活性最好。经液质色谱分析,火炭母水提液中主要成分为酚酸性物质。代表性的酚酸柯里拉京能以时间-剂量依赖性地抑制SiHa细胞增殖。柯里拉京对SiHa细胞的增殖抑制作用在72h时最强,24h的IC_(50)为(42.50±0.38)μmol/L,48h的IC_(50)为(33.20±0.21)μmol/L,72h的IC_(50)为(26.30±0.15)μmol/L。流式细胞检测显示,柯里拉京诱导SiHa细胞的凋亡主要发生在G2阻滞期。结论 火炭母水提液中,以柯里拉京为代表的多羟基酚酸可以显著抑制人宫颈癌SiHa细胞株增殖,是一种潜在的抗宫颈癌活性物质。Objective To analyze the effect on proliferation inhibition of human cervical cancer SiHa cells line by Polygonum sinense in vitro.Methods Polygonum sinense of different solvent extracts was evaluated by Cell Counting Kit-8(CCK8)method processing SiHa cell proliferation activity.Liquid chromatography-mass spectrometry(LC-MS)was used to analyze the Polygonum sinense components in water extraction fluid.CCK8 method was used to determine SiHa cell proliferation activity treated with Corilagin at different concentrations and different processing times,and the IC_(50) was calculated.Corilagin deals with different concentrations after 48 h of SiHa cell apoptosis rate were detected by flow cytometry instrument.Results CCK8 results showed that the water extract of Polygonum sinense was the best in inhibiting the proliferation of SiHa cells,and the liquid mass chromatography analysis showed that the water extract of Polygonum sinense was mainly composed of phenolic acid substances.Corilagin could inhibit SiHa cell proliferation,the strongest proliferation inhibition was 72 h,IC_(50) of 24 h was(42.50±0.38)μmol/L,48 h was(33.20±0.21)μmol/L,72 h was(26.30±0.15)μmol/L.Flow cytometry showed that the apoptosis of SiHa cells induced by corilagin mainly occurred in the G2 block phase.Conclusion Polyhydroxy phenolic acid,represented by corilagin in the water extract of Polygonum sinense,can significantly inhibit the proliferation of human cervical cancer SiHa cell line,and is a potential anti-cervical cancer active substance.
