水稻OsSBEⅡb基因的克隆和时空表达特征及SNP位点多态性

摘　　要：水稻淀粉分支酶(OsSBEⅡb)对水稻支链淀粉的合成至关重要,而水稻支链淀粉的含量与水稻支链淀粉和抗性淀粉的含量密切相关。为了明确OsSBEⅡb基因在水稻淀粉合成中的作用,通过PCR技术扩增获得了湘早籼2号水稻的OsSBEⅡb基因cDNA编码区序列。生物信息学分析表明,OsSBEⅡb基因具有一个编码825个氨基酸多肽的完整读码框;时空表达特征分析发现,该基因表达具有空间特异性,在根、茎、叶、稻壳的表达量很低,主要在稻米胚乳中表达;不同水稻品种OsSBEⅡb基因的编码区存在15个SNP位点变异,表明该基因在不同水稻品种具有丰富的多态性,这些丰富的变异位点为抗性淀粉水稻品种的选育提供了丰富的基因资源。In order to clarify the role of OsSBEⅡb gene in rice starch synthesis,the cDNA coding sequence of OsSBEⅡb gene in Xiangzaoxian 2 was obtained by PCR.Bioinformatics analysis showed that the gene had the maximum open reading frame with 2478 bp and codes a polypeptide containing 825 amino acids.The analysis of temporal-spatial expression characteristics showed that the expression of the gene had spatial specificity:very low level in roots,stems,leaves and rice husk,and very high level in rice endosperm.There were 15 SNP site variations in the coding region of OsSBEⅡb gene in different rice varieties,indicating that the gene has rich polymorphism.The abundant variation sites provide rich gene resources for the breeding of resistant-starch rice variety.
