摘 要:目的探讨骨髓间充质干细胞(BMSCs)干预对大鼠脑胶质瘤C6细胞分化的影响及其相关机制。方法采用Transwell小室共培养骨髓间充质干细胞及C6细胞,并以细胞计数法对C6细胞增值水平进行检测,以流式细胞术对C6细胞的细胞周期进行检测;以免疫荧光对波形蛋白(vimentin)的表达情况进行检测,以免疫组化法对胶质纤维酸性蛋白(GFAP)的表达情况进行检测,以生化法对谷氨酰胺合成酶(GS)活性进行检测。结果BMSCs能够在一定程度上抑制C6细胞增殖;双层培养组C6细胞周期可见GFAP表达明显增强、vimentin表达减弱现象;同时GFAP与vimentin均可见细胞内分布及排列改变现象;同时双层培养组C6细胞内还可见明显得GS活性升高现象。结论骨髓间充质干细胞可诱导C6细胞分化为成熟星形角质细胞。Objective To investigate the effects of bone marrow mesenchymal stem cells(BMSCs)on the differentiation of rat glioma C6 cells and related mechanisms.Methods Transwell cells were used to co-culture BMSCs and C6 cells,and the cell count method was used to detect the level of C6 cell proliferation.The cell cycle of C6 cells was detected by cytometry;immunofluorescence was used to detect vimentin(vimentin).To detect the expression of Glial fibrillary acidic protein(GFAP)by immunohistochemistry,and to detect the activity of glutamine synthetase(GS)by Shenghuafa.Results BMSCs can inhibit the proliferation of C6 cells to a certain extent;the C6 cell cycle of the double-layer culture group showed a significant increase in GFAP expression and a decrease in vimentin expression;at the same time,both GFAP and vimentin showed changes in intracellular distribution and arrangement;while the double-layer culture group In C6 cells,there was also a significant increase in GS activity.Conclusion Bone marrow mesenchymal stem cells can induce C6 cells to differentiate into mature astrocytes.
