防己饮片中生物碱成分的含量测定方法研究

摘　　要：目的:建立高效液相色谱法同时测定防己中木兰花碱、汉防己丙素、防己诺林碱和粉防己碱含量的方法。方法:采用Diamonsil Plus C_(18)色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.3%磷酸溶液,梯度洗脱,流速1.0 mL·min^(-1),检测波长282 nm,柱温30.0℃。结果:木兰花碱、汉防己丙素、防己诺林碱和粉防己碱线性范围分别为2.927~46.834μg·mL^(-1)(r=0.99990)、13.912~278.247μg·mL^(-1)(r=0.99990)、18.471~295.529μg·mL^(-1)(r=0.99935)、21.205~339.277μg·mL^(-1)(r=0.99975);平均回收率分别为100.09%、99.78%、98.21%、100.31%,RSD分别为0.89%、0.92%、1.29%、1.46%。测定8批样品,结果木兰花碱的含量为0.0444%~0.0734%、汉防己丙素的含量为0.6492%~1.0129%、防己诺林碱的含量为0.7437%~0.8973%、粉防己碱的含量为1.2977%~1.9887%。结论:该方法可作为防己饮片的质量控制方法。Objective:A HPLC method was established to simultaneously determine the contents of magnoflorine,cyclanoline,fangchinoline and tetrandrine in Stephaniae Tetrandrae Radix.Methods:A Diamonsil Plus C_(18)column(4.6 mm×250 mm,5μm)was used,the mobile phase was acetonitrile-0.3%phosphoric acid solution gradient e-lution,the flow rate was 1.0 mL·min^(-1),the detection wavelength was 282 nm,and the column temperature was30.0℃.Results:The linear ranges of magnoflorine,cyclanoline,fangchinoline and tetrandrine are 2.927-46.834μg·mL^(-1)(r=0.99990),13.912-278.247μg·mL^(-1)(r=0.99990),respectively,18.471-295.529μg·mL^(-1)(r=0.99935),21.205-339.277μg·mL^(-1)(r=0.99975);the average recoveries were 100.09%,99.78%,98.21%,100.31%,and the RSD were 0.89%,0.92%,1.29%,1.46%.8 batches of samples weretested,and the results showed that the content of magnoflorine was 0.0444%-0.0734%,the content of cyclano-line was 0.6492%-1.0129%,the content of fangchinoline was 0.7437%-0.8973%,and the content of tetran-drine was 1.2977%-1.9887%.Conclusion:The method can be used as a quality control method for StephaniaeTetrandrae Radix decoction pieces.
