摘 要:目的探讨间充质干细胞分泌物对类风湿关节炎细胞因子的调节作用,以期发现有效缓解类风湿关节炎滑膜炎症、减少炎症细胞因子产生的治疗方法。方法随机选取49例类风湿关节炎患者作为研究对象,收集患者流行病学信息资料,采用ELISA方法检测间充质干细胞分泌物刺激下的患者外周血淋巴细胞分泌的TNF-α、IL-1β、IL-6、IL-8的抗体水平;采用QPCR方法检测间充质干细胞分泌物刺激下的类风湿关节炎患者外周血中TNF-α、IL-1β、IL-6、IL-8基因表达水平。结果间充质干细胞分泌物可显著降低类风湿关节炎患者血清中TNF-α抗体水平,IL-1β、IL-6、IL-8的抗体水平也有所降低;间充质干细胞分泌物与类风湿关节炎患者外周血单核细胞共培养24 h后可显著降低细胞中TNF-α、IL-1β、IL-6、IL-8的基因表达水平(P<0.05)。结论间充质干细胞分泌物对类风湿关节炎患者细胞因子具有调节作用。Objective To investigate the regulatory effect of mesenchymal stem cell secretions on cytokines in patients with rheumatoid arthritis(RA),in order to identify an effective way to alleviate the synovial inflammation of rheumatoid arthritis and reduce the production of inflammatory cytokines.Methods 49 patients with rheumatoid arthritis were randomly selected as the research objects,and epidemiological information of these patients was collected.The antibody levels of TNF-α,IL-1,IL-6 and IL-8 secreted by peripheral blood lymphocytes of patients stimulated by mesenchymal stem cell secretion were detected by ELISA,and the gene expression level of TNF-α,IL-1,IL-6,and IL-8 in peripheral blood of patients stimulated by mesenchymal stem cell secretion was detected by qPCR.Results The secretion of mesenchymal stem cells could significantly reduce the serum TNF-αantibody level of patients with rheumatoid arthritis,and the antibody levels of IL-1,IL-6 and IL-8 were also decreased;The gene expression levels of TNF-α,IL-1,IL-6 and IL-8 in the peripheral blood mononuclear cells of patients with rheumatoid arthritis were significantly decreased after co-culture of mesenchymal stem cells secretion and peripheral blood mononuclear cells of patients with rheumatoid arthritis for 24 hours(P<0.05).Conclusion The secretion of mesenchymal stem cells can regulate cytokines in patients with rheumatoid arthritis.
