摘 要:目的:探究银杏叶提取物(GBE)对糖尿病肾病(DN)大鼠肾功能、心功能、炎性反应、β2微球蛋白(β2-MG)及视黄醇结合蛋白4(RBP4)水平的影响。方法:选取36只8周龄的雄性大鼠随机分为对照组、糖尿病肾病模型组(DN组)及银杏叶提取物干预组(DN+GBE组),每组12只。采用一次性腹腔注射链脲佐菌素(60 mg/kg)制作DN模型,分别于干预第4、8、12周抽取大鼠主动脉血备测。比较3组大鼠肾功能[24 h尿蛋白、尿素氮(BUN)、肌酐(Cr)]、心功能[左室射血分数(LVEF)、左心室短轴缩短率(LVFS)]、炎性介质[白介素-6(IL-6)、白介素-8(IL-8)、肿瘤坏死因子(TNF-α)]、β2-MG及RBP4水平差异。结果:组间比较,干预4、8、12周时3组大鼠的24 h尿蛋白、BUN、Cr、IL-6、TNF-α、LVEF、LVFS、β2-MG、RBP4水平差异均具有统计学意义(P<0.05),对照组24 h尿蛋白、BUN、Cr、IL-6、TNF-α、β2-MG、RBP4水平低于DN+GBE组(P<0.05),DN+GBE组24 h尿蛋白、BUN、Cr、IL-6、TNF-α、β2-MG、RBP4水平低于DN组(P<0.05);对照组LVEF、LVFS高于DN+GBE组(P<0.05),DN+GBE组LVEF、LVFS值高于DN组(P<0.05)。结论:银杏叶提取物可有效改善DN大鼠肾功能及心功能,减轻大鼠肾小管损伤,减少炎症因子释放。Objective:To investigate the effects of ginkgo biloba extract(GBE)on renal function,cardiac function,inflammatory response,β2 microglobulin(β2-MG)and retinol binding protein 4(RBP4)in rats with diabetic nephropathy(DN).Methods:36 male rats of 8-week-old were randomly divided into the control group,the model group and the ginkgo biloba extract intervention group,with 12 rats in each group.The DN model was made by one-time intraperitoneal injection of streptozotocin(60 mg/kg),and the aorta blood of the rats was collected after 4 weeks,8 weeks and 12 weeks of interventions.24h urinary protein,urea nitrogen(BUN)and creatinine(Cr)of renal function,left ventricular ejection fraction(LVEF)and left ventricular short axis shortened rate(LVFS)of cardiac function,interleukin-6(IL-6),interleukin-8(IL-8)and tumor necrosis factor(TNF-α)of inflammatory mediators,as well asβ2-MG and RBP4 were compared among the three groups after different time of interventions.Results:There were statistical differences in 24h urinary protein,BUN,Cr,IL-6,TNF-α,LVEF,LVFS,β2-MG and RBP4 among the three groups after 4 weeks,8 weeks and 12 weeks of interventions(P<0.05).In terms of the levels of 24h urinary protein,BUN,Cr,IL-6,TNF-α,β2-MG and RBP4,they were lower in the control group than those in the intervention group(P<0.05),they were lower in the intervention group than those in model group(P<0.05).In terms of the values of LVEF and LVFS,they were higher in the control group than those in the intervention group(P<0.05),and they were higher in the intervention group than those in the model group(P<0.05).Conclusion:Ginkgo biloba extract can effectively improve the renal function and the cardiac function of the DN rats,and it can reduce the injury of renal tubules and the release of inflammatory factors.
