摘 要:以桂花叶为原料,选择乙醇-硫酸铵双水相体系辅助内部沸腾法提取桂花叶中的黄酮,利用单因素试验及响应面模型优化硫酸铵浓度、提取温度,提取时间等提取工艺条件,并与内部沸腾法、双水相萃取法和有机溶剂直接浸提法进行比较。结果表明,桂花叶黄酮的最佳提取条件为硫酸铵浓度0.35 g/mL,提取温度50℃,提取时间20 min,此条件下的黄酮得率平均值为2.534%。此最佳提取条件下的黄酮得率高于相同条件下内部沸腾法、双水相萃取法及有机溶剂直接浸提法的黄酮得率,体现了双水相萃取法和内部沸腾法的协同作用。Flavonoid was extracted from osmanthus leaves as the raw material by and ethanol-ammonium sulfate aqueous two-phase system assisted inner ebullition method.The technological conditions of extraction,includ-ing ammonium sulfate concentration,extraction temperature,and extraction time,were optimized by single-factor experiments and a response surface model.The method was compared with inner ebullition method,aqueous two-phase extraction method,and direct extraction of organic solvent.The optimal extraction conditions of flavonoid in osmanthus leaves were an ammonium sulfate concentration of 0.35 g/mL,extraction temperature of 50℃,and extraction time of 20 min.The average flavonoid yield was 2.534%.The optimized yield of flavonoid was higher than the yields obtained by the inner ebullition method,aqueous two-phase extraction method,and by direct extraction of organic solvent method in the same conditions.The results indicated the synergistic effect between aqueous two-phase method and inner ebullition method.
