摘  要:目的探讨circ_0007897通过调控程序性细胞死亡因子4(PDCD4)介导缺血性心肌损伤的机制。方法分离原代C57小鼠心肌细胞,并分为空白组、缺氧复氧组、缺氧复氧+circ_con组、缺氧复氧+circ_0007897组。检测各组细胞活力、细胞凋亡、细胞乳酸脱氢酶(LDH)水平及细胞内丙二醛(MDA)含量、circ_0007897表达情况及PDCD4蛋白表达情况。结果与空白组相比,缺氧复氧组的细胞活力明显下降,细胞凋亡明显升高,LDH水平及细胞内MDA明显升高,circ_0007897表达水平明显下降,PDCD4的表达明显升高,差异具有统计学意义(q分别=13.86、40.78、14.45、8.12、8.91、9.70,P均<0.05);与缺氧复氧组相比,缺氧复氧+circ_0007897组的细胞活力明显升高,细胞凋亡明显下降,LDH水平及细胞内MDA明显下降,circ_0007897表达水平明显升高,PDCD4表达水平明显下降,差异具有统计学意义(q分别=9.68、20.96、9.13、8.18、14.84、6.51,P均<0.05)。结论circ_0007897对心肌细胞有明显的保护作用,而这种保护作用是通过下调PDCD4来进行调控的。Objective To investigate the mechanism of circ_0007897 mediated ischemic myocardial injury through the regulation of programmed cell death 4(PDCD4).Methods The cardiomyocytes from primary C57 mouse were isolated.They were divided into four groups:blank group,hypoxia reoxygenation group,hypoxia reoxygenation+circ_con group,and hypoxia reoxygenation+circ_0007897 group.Cell viability,apoptosis,LDH level,MDA content,circ_0007897 expression and PDCD4 protein expression were detected.Results Compared with the blank group,the cell viability of the hypoxia reoxygenation group was significantly decreased,cell apoptosis was significantly increased as well as LDH and intracellu-lar MDA level,the expression of circ_0007897 was significantly decreased while the expression of PDCD4 was significant-ly increased(q=13.86,40.78,14.45,8.12,8.91,9.70,P<0.05).When compared with the hypoxia-reoxygenation group,the cell viability of the hypoxia reoxygenation+circ_0007897 group was significantly increased,the cell apoptosis,LDH,and intracellular MDA level was significantly decreased.the expression of circ_0007897 was significantly increased,while the expression of PDCD4 was significantly decreased(q=9.68,20.96,9.13,8.18,14.84,6.51,P<0.05).Conclusion circ_0007897 has obvious protective effect on myocardial cells through downregulating PDCD4.
