摘 要:目的探寻肺不藏魄型失眠大鼠与对照组大鼠相关脏器中(肺、心、肝、脾、肾、大肠、脑)食欲素受体的表达差异,以充实客观实验依据,丰富中医不寐五神分型诊断法的应用。方法随机将16只大鼠分为对照组与模型组,每组8只。通过小平台水环境法制作失眠大鼠模型,运用免疫组化法观察大鼠肝、心、脾、肺、肾、大肠及脑组织中食欲素受体(OX1_(R)、OX2_(R))的表达。结果 (1)模型组较对照组大鼠肺脏组织中OX2_(R)表达上调,心脏组织中OX1_(R)表达上调,脑组织中OX1_(R)、OX2_(R)表达均上调,差异有统计学意义(P<0.01),脾组织中OX1_(R)、OX2_(R)的表达较对照组差异无统计学意义(P>0.01)_(R)(2)两组大鼠肝、肾、大肠组织中OX1_(R)与OX2_(R)均未见明显表达。结论失眠模型大鼠的脑组织中OX1_(R)、OX2_(R)表达均上调,肺脏组织中OX2_(R)表达上调,心脏组织中OX1R表达上调_(R)食欲素受体可能是肺不藏魄型失眠模型大鼠的特征性神经递质之一。Objective The aim is to explore the expression differences of orexin receptors in related organs(lung,heart,liver,spleen,kidney,large intestine and brain)between insomnia rats with lung storing no inferior spirit and control rats,so as to enrich the objective experimental basis and the application of five emotion-based classification and diagnosis of insomnia in traditional Chinese medicine.Methods Sixteen rats were randomly divided into the control group and model group,8 rats in each group.The insomnia rat model was made by small platform-water environment method.The expressions of orexin receptors(OX1_(R) and 0X2_(R))in liver,heart,spleen,lung,kidney,large intestine and brain were detected by immunohistochemistry.Results Compared with the control group,the expressions of 0X2_(R) in lung tissue,OX1_(R) in heart tissue and OX 1_(R) and 0X2_(R) in brain tissue were significantly up-regulated in the model group,with statistically significant differences(P<O.O1),but there were no statistically significant differences in the expressions of OX1_(R)and OX2_(R) in spleen tissue between the control group and model group(P>0.01).②There were no significant expressions of OX1_(R) and OX2_(R) in liver,kidney and large intestine tissues between the two groups.Conclusion The expressions of OX1_(R) and OX2_(R) in brain tissue,OX2_(R) in lung tissue and OX1_(R) in heart tissue of insomnia model rats are up-regulated,and orexin receptors may be one of the characteristic neurotransmitters in insomnia rats with lung storing no inferior spirit.
