摘 要:花秆孝顺竹(Bambusa multiplex f.alphonso-karri)作为最具抗旱和耐寒能力的丛生型观赏竹,其独有的"孝"文化内涵,在竹文化传播与传承中具有重要意义。为发挥竹组培苗在居家美化、庭园绿化特别是竹盆栽配植中的优势,探索花秆孝顺竹不定芽诱导增殖、生根诱导的最佳培养基配方,建立高效的组培快繁体系,文章以花秆孝顺竹当年生枝条的带芽茎段为外植体,研究了6-苄基腺膘呤(6-BA)、噻二唑苯基脲(TDZ)单因素及其组合对花秆孝顺竹不定芽诱导增殖的影响,以及萘乙酸(NAA)和6-BA组合对花秆孝顺竹生根诱导的影响。结果表明:在不定芽诱导增殖试验中,单独使用TDZ控制褐化优于单独使用6-BA;2种调节剂组合使用其增殖效果优于单一调节剂,在6-BA 3.00 mg/L+TDZ 0.05 mg/L配比下,外植体不定芽的诱导数最多,为4.80个。在生根试验中,0.30 mg/L的6-BA与NAA搭配可以促进花秆孝顺竹组培苗生根,其中0.10 mg/L NAA+0.30 mg/L 6-BA处理的组培苗生根表现最优,生根率达100%,平均根数为1.70条,平均根长为1.30 cm。研究所建立的花秆孝顺竹组培快繁体系可为其种苗大规模生产提供技术支撑。Bambusa multiplex f. alphonso-karri is the most drought resistant and cold resistant ornamental sympodial bamboo. It’s unique cultural connotation of "filial piety" is of great significance in the dissemination and inheritance of bamboo culture. In order to give full play to the advantages of the bamboo seedlings from tissue culture in home beautification, gardening, and especially pot planting, and explore its best medium formula for adventitious bud induction, proliferation and rooting induction of bamboo for efficient in vitro rapid propagation system, this paper uses the bamboo stem segments with buds in internodes of current year branches of B. multiplex f. alphonso-karri as explants to study the effects of 6-BA, TDZ single factor and their combination on adventitious bud induction and proliferation of B. multiplex and the effects of NAA and 6-BA combination on its rooting induction. The results show that, in the experiment of adventitious bud induction and proliferation, TDZ used alone is better than 6-BA used alone in browning control;the proliferation effect of the combination of the two regulators is better than that of 6-BA alone, and the average number of buds is the highest, reaching 4.8 at the ratio of 6-BA 3 mg/L and TDZ 0.05 mg/L. In the rooting test, 6-BA at 0.3 mg/L and NAA have better rooting performance, among which NAA 0.1 mg/L + 6-BA 0.3 mg/L have the best rooting performance, with rooting rate of 100%, average root number at 1.70 and average root length at 1.30 cm. The tissue culture and rapid propagation system established in this study can provide technical support for large-scale seedling production of B. multiplex.
