摘 要:目的:探讨加味小柴胡汤对哮喘模型大鼠气道重塑中TGF-β_(1)/Smad3信号传导通路的影响。方法:36只雄性SD大鼠随机分为空白组、模型组、西药组,中药低、高剂量组和中药联合西药组,每组6只。应用卵清白蛋白致敏大鼠哮喘,并制备哮喘模型。哮喘模型建立成功开始,各用药组分别给予同等剂量药物进行灌胃治疗,空白组及模型组分别给予等量生理盐水灌胃治疗。HE染色法观察肺组织气道壁嗜酸性粒细胞浸润;运用图像分析技术测定各组肺内支气管壁面积(Wat)、支气管平滑肌面积(Wam)等指标;免疫组织化学法检测各组肺组织TGF-β_(1)、Smad3蛋白表达水平。结果:与空白组比较,模型组气道壁嗜酸性粒细胞浸润明显增多(P<0.05),大鼠Wat、Wam显著增高(P<0.05);与模型组比较,各给药组气道壁嗜酸性粒细胞浸润明显减少(P<0.05),大鼠Wat、Wam显著降低(P<0.05)。与空白组比较,模型组肺组织TGF-β_(1)、Smad3蛋白表达升高(P<0.05);与模型组比较,各给药组肺组织TGF-β_(1)、Smad3蛋白表达降低(P<0.05)。结论:对于肺组织气道壁嗜酸性细胞浸润以及TGF-β_(1)、Smad3蛋白表达,加味小柴胡汤均能从不程度上降低,从而减缓气道重塑。Objective:To investigate the effect of Jiawei Xiaochaihu decoction on TGF-β_(1)/Smad3 signaling pathway in airway remodeling of asthmatic rats.Methods:Thirty six male SD rats were randomly divided into blank group,model group,western medicine group,low-dose group,high-dose group and Chinese medicine combined with western medicine group,with 6 rats in each group.Asthmatic rats were sensitized with ovalbumin.Asthma model was established successfully,each medication group was given the same dose of drugs for intragastric treatment.Blank group and model group were given the same amount of normal saline intragastric treatment.Eosinophil infiltration in the airway wall was observed by HE staining.The area of bronchial wall(Wat)and the area of bronchial smooth muscle(Wam)were measured by image analysis.The lung tissue TGF-β_(1)and Smad3 protein expression level were detected by immunohistochemistry.Results:Compared with the blank group,the infiltration of eosinophils in the airway wall of the model group was significantly increased(P<0.05).Wat and Wam of rats were significantly higher than that of the blank group(P<0.05),and the expression levels of other drug groups were lower than that of the model group(P<0.05).Compared with the blank group,the expression levels of TGF-β_(1)and Smad3 protein in the lung tissues of the model group were increased(P<0.05),while the expression levels of TGF-β_(1)and Smad3 protein of other drug groups were lower than that of the model group(P<0.05),with statistically significant differences.Conclusion:For eosinophilic cell infiltration and expression of TGF-β_(1)and Smad3 protein in airway wall of lung tissue,Xiaochaihu decoction can reduce the airway remodeling to some extent.
