帕瑞昔布钠对宫颈癌细胞COX2/PGE2通路及细胞生物学行为的影响

摘　　要：目的探讨帕瑞昔布钠对宫颈癌HeLa细胞环氧合酶2/前列腺素E2(COX2/PGE2)通路及增殖、凋亡、侵袭行为的影响。方法体外培养人宫颈癌HeLa细胞,分为对照组(不加药物)、低浓度帕瑞昔布钠组(10μmol/L帕瑞昔布钠)、中浓度帕瑞昔布钠组(40μmol/L帕瑞昔布钠)和高浓度帕瑞昔布钠组(160μmol/L帕瑞昔布钠)。MTT法检测HeLa细胞增殖情况;流式细胞仪检测HeLa细胞凋亡情况;Transwell法检测HeLa细胞侵袭情况;实时荧光定量PCR(qRT-PCR)法检测HeLa细胞中COX2、PGE2 mRNA表达情况;蛋白免疫印迹(Western blot)法检测HeLa细胞中COX2、PGE2、Bax、MMP-9蛋白表达情况。结果与对照组比较,低、中、高浓度帕瑞昔布钠组HeLa细胞增殖率、细胞中COX2、PGE2 mRNA及蛋白表达水平、MMP-9蛋白表达水平显著降低(P<0.05),细胞凋亡率及细胞中Bax蛋白表达水平显著升高(P<0.05),侵袭细胞数显著减少(P<0.05)。随着帕瑞昔布钠浓度的升高,HeLa细胞增殖率、细胞中COX2、PGE2 mRNA及蛋白表达水平、MMP-9蛋白表达水平依次降低(P<0.05),细胞凋亡率及细胞中Bax蛋白表达水平依次升高(P<0.05),侵袭细胞数依次减少(P<0.05)。结论帕瑞昔布钠可抑制宫颈癌HeLa细胞中COX2、PGE2表达及细胞增殖、侵袭过程,并诱导细胞凋亡。Objective To investigate the effects of parecoxib sodium(PS)on cyclooxygenase-2/prostaglandin E2(COX2/PGE2)pathway,proliferation,apoptosis and invasion of cervical cancer HeLa cells.Methods Human cervical cancer HeLa cells were cultured in vitro and divided into control group(without drug treatment),low-dose PS group(10μmol/L),medium-dose PS group(40μmol/L)and high-dose PS group(160μmol/L).MTT assay was used to detect the proliferation of HeLa cells,and the cell apoptosis was detected by flow cytometry.Transwell assay was used to detect the invasion of HeLa cells,and real-time fluorescence quantitative PCR(qRT-PCR)was used to detect the mRNA expression of COX2 and PGE2,and Western Blot was used to detect the expression levels of COX2,PGE2,Bax and MMP-9 in HeLa cells.Results Compared with those in control group,the proliferation rate of HeLa cells,the expression levels of COX2 and PGE2 mRNA and protein as well as MMP-9 were significantly decreased in low-dose,medium-dose and high-dose parecoxib sodium groups(P<0.05),however,the apoptosis rate and the expression level of Bax protein were significantly increased(P<0.05),and the number of invasive cells was significantly decreased(P<0.05).With the increase of parecoxib sodium concentration,the proliferation rate of HeLa cells,and the expression levels of COX2 and PGE2 mRNA and protein as well as MMP-9 were significantly decreased in order of low-dose,medium-dose and high-dose parecoxib sodium groups(P<0.05),however,the apoptosis rate and expression levels of Bax protein were significantly increased(P<0.05),and the number of invasive cells were significantly decreased in this order(P<0.05).Conclusion Parecoxib sodium can inhibit the expression of COX2,PGE2,proliferation and invasion of cervical cancer HeLa cells,moreover,which can induce cell apoptosis.
