摘  要:目的分析miR-149-5p过表达对重症肺炎大鼠肺组织PI3K/Akt/mTOR通路的影响。方法将大鼠肺细胞分为对照组、下调组和上调组,并通过细胞转染建立稳定转染的下调组和上调组。采用免疫组织化学法检测凋亡蛋白表达量,采用酶联免疫法检测大鼠炎性因子水平,采用MTT比色法检测GSH、ROS及T-AOC含量,采用实时荧光定量PCR测定各组细胞中PI3KAktmTOR通路因子表达量。结果过表达组Caspase-3、Caspase-8、Caspase-9蛋白表达量低于沉默组,差异有统计学意义(P<0.05);过表达组大鼠肺组织中炎性因子水平低于沉默组,差异有统计学意义(P<0.05);过表达组GSH、ROS低于沉默组,T-AOC含量高于沉默组,差异有统计学意义(P<0.05);过表达组Bax mRNA表达量高于沉默组,Bcl-2、BDNF、pERK、VGF mRNA表达量低于沉默组,差异有统计学意义(P<0.05)。结论过表达miR-149-5p,可通过作用于PI3K/Akt/mTOR通路,调控Bax、Bcl-2、BDNF、pERK、VGF mRNA表达量,减轻炎性反应,抑制细胞凋亡。Objective To investigate the effects of miR-149-5p overexpression on the PI3K/Akt/mTOR pathway in the lung tissue of rats with severe pneumonia.Methods The lung cells of rats were divided into control group,down-regulation group and up-regulation group,and the stable transfection in down-regulation group and up-regulation group was established by cell transfection.Immunohistochemistry was used to detect the expression levels of apoptosis protein,and enzyme-linked immunoassay was used to detect the level of inflammatory factors in rats,MTT colorimetric method was used to detect the contents of GSH,ROS and T-AOC,and real-time fluorescence quantitative PCR was used to detect the expression levels of cells PI3KAktmTOR pathway factor.Results The expression levels of Caspase-3,Caspase-8,and Caspase-9 protein in overexpression group were significantly lower than those in silencing group(P<0.05),and the levels of inflammatory factors as well as the expression levels of GSH and ROS in overexpression group were significantly lower than those in silencing group(P<0.05),however,the levels of T-AOC were significantly higher than those in silencing group(P<0.05).Moreover the expression levels of Bcl-2,BDNF,pERK,VGF mRNA in overexpression group were significantly lower than those in silencing group(P<0.05).Conclusion The overexpression of miR-149-5p can regulate the expression levels of Bax,Bcl-2,BDNF,pERK,and VGF mRNA via PI3K/Akt/mTOR pathway,so as to relieve inflammation reaction and inhibit cell apoptosis.
