摘 要:目的探讨猴头健胃灵片治疗胃黏膜损伤的可能作用机制。方法30只SD大鼠随机分为空白组、模型组和猴头健胃灵组,每组10只。模型组和猴头健胃灵组采用冰乙酸注射法制备胃黏膜损伤大鼠模型。造模后猴头健胃灵组予浓度为24 mg/ml的猴头健胃灵片水溶液1 ml/100 g灌胃,空白组和模型组给予等体积纯水灌胃。7天后比较各组大鼠体重及24 h进食量,测量黏膜损伤面积并计算黏膜损伤愈合率,HE染色和AB-PAS染色观察大鼠胃黏膜组织病理变化,检测胃黏膜及血清前列腺素E_(2)(PGE_(2))含量和胃黏膜组织环氧合酶2(COX-2)蛋白表达水平。结果与空白组比较,模型组大鼠体重和24 h进食量减少,血清、胃黏膜PGE_(2)含量降低,胃黏膜COX-2蛋白水平升高(P<0.05或P<0.01);与模型组比较,猴头健胃灵组大鼠24 h进食量增加,溃疡面积缩小,溃疡愈合率升高,血清、胃黏膜PGE_(2)含量升高,胃黏膜COX-2蛋白水平降低(P<0.05或P<0.01)。模型组大鼠在造模术后1天时,胃黏膜胃腺结构紊乱、部分丢失,出现了大量黏膜下水肿、炎症细胞浸润和散在出血点,术后1周时,黏膜损伤有所恢复,炎症、出血有所减少;猴头健胃灵组大鼠大部分胃黏膜光滑完整,未见明显的胃腺结构紊乱或丢失。结论猴头健胃灵片具有胃黏膜保护作用,其机制可能是通过促进胃黏膜合成和分泌PGE_(2)、抑制COX-2的表达,从而保护胃黏膜屏障。Objective To explore the possible mechanism of Houtou Jianweiling Tablets(猴头健胃灵片,HJT)in treatment of gastric mucosal injury(GMI).Methods Thirty SD rats were randomly divided into blank group,model group and HJT group,with 10 rats in each group.Rats in the model group and the HJT group were injected with acetic acid to establish the GMI model.After modeling,the HJT group was given 1 ml/100 g water solution of HJT at concentration of 24 mg/ml by gavage,while the blank group and the model group received equal volume of pure water by gavage.After seven days,the weight of rats and 24 h dietary intake were assessed,the area of gastric mucosal injury was measured,and healing rate of the injury were calculated.HE staining and AB-PAS staining were used to observe the pathological changes of gastric mucosa in rats;the prostaglandin E2(PGE2)content in the gastric mucosa and serum,and the cyclooxygenase 2(COX-2)protein expression level were detected.Results Compared to the blank group,the model group had decreased rats weight and 24 h dietary intake,reduced PGE2 content in the serum and gastric mucosa,as well as elevated COX-2 protein level in gastric mucosa(P<0.05 or P<0.01).Com⁃pared to those in the model group,the 24 h dietary intake in the HJT group increased,the ulcer area decreased,the healing rate of the injury increased,the PGE2 content in the serum and gastric mucosa increased,the COX-2 protein level in gastric mucosa increased(P<0.05 or P<0.01).One day after the model was successfully established,the structure of the gastric mucosa in the model group was disordered and partially lost,and there was a large amount of submucosal edema,inflammatory cell infiltration and scattered bleeding points;one week after the modelling,the mucosal injury was restored to certain degree,and the inflammation and bleeding were reduced.Most of the gas⁃tric mucosa in the HJT group was smooth and intact,and there was no obvious structural disorder or loss of gas⁃tric glands.Conclusion HJT can protect the gastric mucosa,
