摘　　要：以多色荧光标记引物, PCR复合扩增多个短串联重复序列(short tandem repeat, STR)基因座的DNA检验技术是检测法医各类生物学检材的主要方法。但陈旧生物学检材中的血红素、胶原蛋白及腐败产生的鞣酸、白骨化残骸中的钙离子、环境中的腐殖酸及衣物中的靛蓝等染料均可作为抑制剂影响其对模板DNA的PCR扩增。摸清PCR抑制剂的种类和抑制机制, 通过增减模板DNA、Taq DNA聚合酶及在PCR缓冲体系中添加相对应的抑制剂拮抗剂, 提高PCR反应的成功率, 对积案的侦破及其他遗传标记的检测均有重要意义。Using multicolor fluorescently labeled primers,PCR composite amplification of multiple short tandem repeat(STR)loci DNA testing technology is the main method for detecting various biological samples of forensic medicine.However,dyes such as heme,collagen and tannic acid produced by corruption in old biological samples,calcium ions in white bones,humic acid in the environment,and indigo in clothing can all be used as inhibitors to affect PCR amplification.Find out the types and inhibition mechanisms of PCR inhibitors,increase or decrease template DNA,Taq DNA polymerase,and add corresponding inhibitor antagonists to the PCR buffer system to increase the success rate of the PCR reaction.It is of great significance to the detection of long-standing cases and the detection of other genetic markers.
