摘 要:本研究旨在从临床仔猪腹泻样品中分离猪流行性腹泻病毒(PEDV),并对其S基因进行测序分析。对腹泻仔猪小肠样品进行RT-PCR检测、病毒的分离培养、RT-PCR和间接免疫荧光鉴定、S基因测序及遗传演化分析。结果显示:仔猪腹泻由流行性腹泻病毒引起;在Vero细胞上成功分离流行性腹泻病毒,命名为XP2018,该毒株细胞病变明显,并且连续传代细胞病变稳定;该毒株S基因全长为4161 bp,与中国经典毒株CV777 S基因核苷酸同源性为92.6%,氨基酸同源性为91.0%;与近年来中国分离的其他毒株S基因核苷酸同源性为96.9%~97.6%,氨基酸同源性为96.9%~98.0%;与美国参考毒株S基因核苷酸同源性为96.9%~97.6%,氨基酸同源性为96.6%~98.3%;遗传进化分析显示XP2018毒株与与目前中国流行毒株在同一分支,与经典毒株CV777不在一个分支上。本研究成功分离1株猪流行性腹泻病毒流行毒株,为PEDV疫苗研发奠定了基础。The objective of this study was to isolate Porcine epidemic diarrhea virus(PEDV)from intestinal samples of diarrhea piglets and analyze the S gene of the isolated strain.The intestinal samples of diarrhea piglets were inoculated onto vero cells and the isolated virus strain XP2018 was examined in RT-PCR and IFA and its S gene was sequenced for phylogenetic analysis.The XP2018 strain caused visible CPE on Vero cells.The full length of its S gene was 4161 bp.Sequence analysis showed that the XP2018 strain had the nucleotide/amino acid homology of 92.6%/91.0%,96.9-97.6%/96.9%-98.0%and 96.9%-97.6%/96.6%-98.3%with the reference Chinese strain CV777,other Chinese strains isolated in recent years and American strains,respectively.The phylogenetic analysis indicated that the XP2018 strain was clustered into the same group with the recently isolated Chinese strains but far from the reference Chinese strain CV777.In summary,this study laid the foundation for the development of PEDV vaccine.
