七彩山鸡内源性禽白血病病毒的鉴定及其env基因序列分析

摘　　要：为了解七彩山鸡感染内源性禽白血病病毒(ALV)情况,从上海某七彩山鸡场采集200份血浆样品,经DF1细胞分离和ALV p27抗原ELISA方法检测出13份阳性样品。将含有阳性样品的第一代细胞裂解液及细胞上清液同时盲传接种至第2代细胞,培养9 d后,p27抗原均未被检测到。对ALV p27抗原阳性细胞培养物进行env基因的扩增,最终得到了1株内源性ALV前病毒序列,其基因序列与已知的禽白血病病毒同源性为39.1%~66.7%。以上结果表明,该七彩山鸡品系中存在内源性禽白血病病毒感染。In order to investigate the infection of Endogenous avian leukosis virus in Phasianus colchicas,200 plasma samples were collected from a Phasianus colchicas farm in Shanghai 13 samples were positive by virus isolation and antigen p27 ELISA test.Both positive cell and supernatant were cultured by cell blind passage,and the result was negative by antigen p27 ELISA test 9 days later.The env gene was amplifi ed and sequenced from p27 antigen positive cell culture.Genomic analysis showed the homology was 39.1%~66.7%between the gene sequence and ALV.The results showed that Endogenous avian leukemia virus was existing in the Phasianus colchicas,which could interfere with the antigen p27 ELISA test.
