黄腐酸钠预处理对大鼠心肌缺血再灌注损伤的作用及其机制研究

摘　　要：目的:观察黄腐酸钠预处理对大鼠心肌缺血再灌注损伤的作用,并阐明相关机制。方法:将SD大鼠随机分为假手术组,缺血再灌注(I/R)模型组,黄腐酸钠高、中、低剂量组(150、100、50 mg/kg)。观察大鼠心肌形态学改变,检测心肌梗死面积,测定相关血清指标水平。结果:与假手术组比较,I/R模型组心肌形态学及梗死面积等,差异有统计学意义(P<0.05);与I/R模型组比较,三个剂量组的黄腐酸钠预处理均能使大鼠心肌梗死面积减小;肌酸激酶同工酶(CK-MB)、肌钙蛋白T(cTnT)含量下降;超氧化物歧化酶(SOD)活性提高;肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、丙二醛(MDA)水平降低。结论:黄腐酸钠能够限制氧化应激,抑制炎性反应,对心肌缺血再灌注损伤具有良好的保护效果。Objective:To observe the effect of sodium fulvic acid pretreatment on myocardial ischemia-reperfusion injury in rats and clarify the related mechanism.Methods:SD rats were randomly divided into sham operation group,ischemia-reperfusion(I/R)model group and sodium fulvic acid high,medium and low dose groups(150,100 and 50 mg/kg).The morphological changes of rat myocardium were observed,the infarct size and the level of related serum indexes were measured.Results:Compared with the sham operation group,there were significant differences in myocardial morphology and infarct size in the I/R model group(P<0.05).Compared with the I/R model group,the pretreatment of sodium fulvic acid in the three dose groups could reduce the myocardial infarction size of rats;The contents of creatine kinase isoenzyme(CK-MB)and cardiac troponin T(cTnT)decreased;superoxide dismutase(SOD)activity increased;tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and malondialdehyde(MDA)levels decreased.Conclusion:Sodium fulvic acid can limit oxidative stress,inhibit inflammatory response and have a good protective effect on myocardial ischemia-reperfusion injury.
