摘 要:为构建山东省地方桃种质资源分子身份证,本研究以60份山东省地方桃品种、名优特品种、选育品种为材料,从桃种质的SSR富集文库中开发并设计30对引物,选出10对具有良好多态性的引物进行PCR扩增,采用SSR荧光标记毛细管电泳技术检测扩增条带分子质量的方法进行带型分析,采用阿拉伯数字1~9和小写英文字母对不同带型进行编码,无带编码为0,按照10对引物扩增带型数由少到多的顺序将编码串联,构建供试样品的分子身份证。结果表明,10对SSR引物共检测到58个SSR等位位点和116个带型,平均每对引物扩增的等位位点为5.8个,带型11.6个;多态性信息含量变幅为0.2044~0.7378,平均0.5699,说明引物的多态性较高,能够有效揭示60份桃种质的遗传多样性。通过对扩增带型的分析并编码,成功构建了60份桃种质的分子身份证。遗传聚类分析的结果与分子身份证结果一致。本研究结果可为山东省桃种质资源的保护和利用奠定基础,对于桃产业的发展具有重要意义。Sixty peach germplasm resources collected from different regions of Shandong Province were used as materials to establish the molecular ID with simple sequence repeat(SSR)markers.Thirty pairs of primers were developed and designed from SSR-enriched library,and 10 pairs with high polymorphisms and good repeatability were selected and used for amplification and capillary electrophoresis.The products of the 60 samples with each primer pairs were analyzed.Each band pattern was coded by different single digit or lowercase letters.Meanwhile,according to the number of band patterns,in an ascending order,the 10 primer pairs were decided,and molecular ID codes of the 60 peach germplasms were established.The results showed that a total of 58 polymorphic alleles and 116 polymorphic patterns were revealed using the 10 primer pairs with the means of 5.8 alleles and 11.6 patterns for each primer pairs;the polymorphic information content(PIC)ranged from 0.2044 to 0.7378 with an average value of 0.5699,so the 10 pairs of SSR primers with high polymorphism could evaluate the genetic diversity of the 60 peach germplasms effectively.Using the combination of single digits and lowercase English letters to encode different band types,molecular ID of the 60 Shandong peach germplasms were successfully constructed.The cluster results basically coincided with the molecular ID codes.The results could provide references for variety identification,evaluation and utilization,and had great significance to development of peach industry.
