摘 要:为了解苹果植株不同部位内生固氮菌的群落结构差异,本研究以苹果营养系砧木M9T337为材料,基于NifH基因采用Illumina Miseq测序技术,对苹果砧木M9T337根、枝、叶内生固氮菌的群落组成和多样性进行分析。结果表明,苹果砧木M9T337各组织部位均有丰富的内生固氮菌,不同部位内生固氮菌群的组成及丰度存在差异。苹果砧木3个部位内生固氮菌除丰富度指数外,Alpha多样性指数无显著性差异。Beta多样性分析结果表明根部与枝部内生固氮菌群落结构差异较大。在门水平上,变形菌门(Proteobacteria)是根、枝、叶部位的优势菌门。在属水平上,慢生根瘤菌属(Bradyrhizobium)是叶部和枝部的优势菌属,固氮螺菌属(Azospirillum)是根部的优势菌属。根部有2个特有属,枝部有2个特有属,叶部有4个特有属。此外,本研究首次发现广古菌门(Euryarchaeota)内生固氮古菌为叶部所特有。In order to know about the differences in community structure of endophytic nitrogen-fixing bacteria in different parts of apple rootstock M9 T337,the Illumina Miseq sequencing technology was used based on NifH gene to analyze the community composition and diversity of endophytic nitrogen-fixing bacteria in roots,branches and leaves.The results showed that there were abundant endogenous nitrogen-fixing bacteria in the three tissues of apple stock M9 T337,and there were differences in the composition and abundance among different tissues.There was no significant difference in alpha diversity index of endophytic nitrogen-fixing bacteria among the three parts of apple rootstock,but there was significant difference in richness index.The beta diversity analysis showed that the community structure of endophytic nitrogen-fixing bacteria was different obviously between root and branch groups.At the phylum level,the Proteobacteria was the dominant phylum in roots,branches and leaves.At the genus level,Bradyrhizobium was the dominant genus in leaves and branches,and Azospirillum was the dominant genus in roots.There were two endemic genera in roots,two endemic genera in branches,and four endemic genera in leaves.In addition,it was found at the first time that the Euryarchaeota endophytic nitrogen-fixing archaea is unique in the leaves of M9 T337.
