妇科千金方及其拆方、单味饮片主要成分分析及10种成分测定

摘　　要：目的分析妇科千金方(穿心莲、单面针、功劳木等)及其拆方(抗炎方、抑菌方、补益方)、单味饮片主要成分,并测定原儿茶酸、木兰花碱、芦丁、染料木苷、党参炔苷、小檗碱、穿心莲内酯、染料木素、脱水穿心莲内酯、藁本内酯的含量。方法制备全方、拆方、单味饮片,其70%甲醇提取物的HPLC分析采用Kromasil 100-5-C_(18)色谱柱(4.6 mm×250 mm,5μm);流动相乙腈-0.1%甲酸,梯度洗脱;体积流量1.0 mL/min;柱温30℃;检测波长254 nm。结果 10种成分在各自范围内线性关系良好(R^(2)≥0.998 5),平均加样回收率96.51%~102.03%,RSD 0.88%~2.97%。木兰花碱、小檗碱、穿心莲内酯、脱水穿心莲内酯归属于抑菌方,染料木苷、染料木素、芦丁归属于抗炎方,原儿茶酸、党参炔苷、藁本内酯归属于补益方。各成分含量在单味饮片中最高,全方中最低。结论妇科千金方主要成分均可归属于拆方、单味饮片中。该方法稳定可靠,可为研究妇科千金方质量控制及配伍规律提供参考。AIM To analysis the main components in Gynecological Qianjin Formula[(Andrographis paniculata(Burm.f.) Nees,Zanthoxylum dissitum Hemsley,Mahonia bealei(Fort.)Carr.,etc.] and its decomposed formulas(anti-inflammatory formula,bacteriostasis formula,tonifying formula),single decoction pieces,and to determine the contents of protocatechuic acid,magnoflorine,rutin,genistin,lobetyolin,berberine,andrographolide,genistein,dehydrated andrographolide and ligustilide.METHODS Whole formula,decomposed formulas and single decoction pieces were prepared,after which the HPLC analysis of their 70%methanol extracts was prepared on a 30 ℃ thermostatic Kromasil 100-5-C_(18) column(4.6 mm×250 mm,5 μm),with the mobile phase comprising of acetonitrile-0.1%formic acid flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelength was set at 254 nm.RESULTS Ten constituents showed good linear relationships within their own ranges(R^(2)≥0.998 5),whose average recoveries were 96.51%-102.03% with the RSDs of 0.88%-2.97%.Magnoflorine,berberine,andrographolide and dehydrated andrographolide were attributed to bacteriostasis formula,genistin,genistein and rutin were attributed to anti-inflammatory formula,protocatechuic acid,lobetyolin and ligustilide were attributed to tonifying formula.The contents of various constituents were the highest in the single decoction pieces and the lowest in the whole formula.CONCLUSION All the main constituents in Gynecological Qianjin Prescription can be attributed to the decomposed formulas and the single decoction pieces.This stable and reliable method can provide a reference for studying the quality control and compatibility pattern of Gynecological Qianjin Prescription.
