摘 要:目的观察芪箭消瘿方对自身免疫性甲状腺炎(AIT)大鼠甲状腺炎症损伤及TLR4/MyD88/NF-κB信号通路的影响。方法 48只雌性SD大鼠随机分为对照组、模型组、硒酵母组(36μg/kg)及芪箭消瘿方低(1.8 g/kg)、中(3.6 g/kg)、高(7.2 g/kg)剂量组,每组8只。采用高碘水喂养联合皮下注射猪甲状腺球蛋白与弗氏佐剂诱导AIT大鼠模型,造模后连续灌胃给药6周。ELISA检测血清FT3、FT4、TGAb、TPOAb、IFN-γ、TNF-α水平,HE染色观察甲状腺组织形态学变化并进行炎症评分,RT-qPCR及Western blot检测甲状腺TLR4/MyD88/NF-κB信号通路相关mRNA和蛋白表达。结果与模型组比较,芪箭消瘿方各剂量组及硒酵母组大鼠甲状腺炎症损伤改善,炎症评分降低(P<0.01),血清FT3、FT4、TGAb、TPOAb、IFN-γ、TNF-α水平降低(P<0.05,P<0.01),芪箭消瘿方中、高剂量组TLR4/MyD88/NF-κB信号通路相关mRNA及蛋白表达均降低(P<0.01)。结论芪箭消瘿方可能通过调控TLR4/MyD88/NF-κB信号通路,减轻促炎细胞因子表达,改善甲状腺组织炎症损伤,从而起到保护甲状腺的作用。AIM To observe the effects of Qijian Xiaoying Prescription on thyroid inflammatory injury and TLR4/MyD88/NF-κB signaling pathway in rats with autoimmune thyroiditis(AIT).METHODS Forty-eight female SD rats were randomly divided into control group,model group,selenium yeast group(36 μg/kg),and Qijian Xiaoying Prescription groups for low-dose,medium-dose,and high-dose administrations(1.8,3.6,7.2 g/kg),with eight rats in each group.The AIT rat models established by high-iodine water feeding combined with subcutaneous injection of porcine thyroglobulin and Freund’s adjuvant were then intragastrically given the corresponding drugs for six weeks.Subsequently,the rats were subjected to the detection of serum levels of FT3,FT4,TGAb,TPOAb,IFN-γ and TNF-α by ELISA;the observation of the morphological changes of thyroid tissue and inflammation scoring by HE staining;and the detection of the mRNA and protein expressions of TLR4/MyD88/NF-κB signaling pathway in thyroid tissue by RT-qPCR and Western blot.RESULTS Compared with the model group,the groups intervened with selenium yeast and Qijian Xiaoying Prescription shared improved thyroid inflammatory injury and decreased inflammation scores(P<0.01),decreased serum levels of FT3,FT4,TGAb,TPOAb,IFN-γ and TNF-α(P<0.05,P<0.01).The medium-dose and high-dose Qijian Xiaoying Prescription groups displayed reduced mRNA and protein expressions of TLR4/MyD88/NF-κB signaling pathway in thyroid tissue(P<0.01).CONCLUSION Qijian Xiaoying Prescription may reduce the expressions of pro-inflammatory factors to improve thyroid inflammatory injury via regulation of TLR4/MyD88/NF-κB signaling pathway.
