摘 要:目的建立兴安升麻Cimicifuga dahurica(Turcz) Maxim UPLC指纹图谱,并测定4种酚酸类质量标志物。方法兴安升麻30%甲醇提取物的分析采用Agilent Eclipse Plus C_(18)色谱柱(2.1 mm×100 mm,1.8μm);流动相乙腈-0.1%的磷酸溶液;体积流量0.25 mL/min;柱温35℃;检测波长220 nm(指纹图谱)和316 nm(定量分析)。结果 20批样品指纹图谱中有11个共有峰,相似度均≥0.896。阿魏酸、异阿魏酸、升麻酸B和升麻酸F分别在0.105 4~10.54(r=0.999 0)、2.54~254(r=0.999 0)、10.72~1 072(r=0.999 7)、10.26~1 026μg/mL(r=0.999 8)范围内线性关系良好,平均加样回收率(RSD)分别为100.3%(1.34%)、99.3%(1.76%)、99.7%(0.35%)、99.6%(0.53%)。结论该方法稳定可靠,可用于兴安升麻的质量控制。AIM To establish the UPLC fingerprints of Cimicifuga dahurica(Turcz) Maxim and to determine the contents of four phenolic acid quality markers.METHODS The analysis of 30% methanol extract of C.dahurica was developed on 35 ℃ thermostatic Agilent Eclipse plus C_(18)(2.1 mm×100 mm,1.8 μm),with the mobile phase comprising of acetonitrile-0.1% phosphoric acid flowing at 0.25 mL/min in a gradient elution manner,and the detection wavelength was set at 220 nm for fingerprint analysis and 316 nm for quantitative analysis,respectively.RESULTS There were 11 common peaks in the fingerprints of 20 batches of samples,with the similarities of more than 0.896.Ferulic acid,isoferulic acid,cimicifuga B,and cimicifuga F showed good linear relationships within the ranges of 0.105 4-10.54(r=0.999 0)、2.54-254(r=0.999 0)、10.72-1 072(r=0.999 7)、10.26-1 026 μg/mL(r=0.999 8),whose average recoveries were 100.3%,99.3%,99.7%,99.6% with the RSDs of 1.34%、1.76%、0.35%、0.53%,respectively.CONCLUSION This stable and reliable method can be used for the quality control of C.dahurica.
