摘 要:目的探讨普罗布考对高脂血症(HL)大鼠脑缺血再灌注后神经功能、脑梗死比例及核转录因子κB(NF-κB)表达影响。方法选取120只健康雄性SD大鼠,普通喂养1 d后分为普通组(20只),高脂血症组(100只),分别给予普通饲料喂养和高脂饲料喂养。高脂血症组大鼠按照随机数字表法分为假手术组、缺血再灌注组、低剂量普罗布考组、高剂量普罗布考组,各25只大鼠。低剂量普罗布考组、高剂量普罗布考组分别给予100、500 mg/kg灌胃,假手术组及缺血再灌注组给予相同剂量单一0.5%羧甲基纤维素钠灌胃,每日1次,连续2周。除假手术组外,缺血再灌注组、低剂量普罗布考组、高剂量普罗布考组制备大鼠大脑中动脉栓塞模型,以脑缺血2 h再灌注时间点的不同(6、12、24、48、72 h)随机分为5个亚组(每组5只),缺血2 h后再灌注24 h行氯化三苯基四氮唑(TTC)染色计算脑梗死体积比;检测血清中总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)水平,评估各组大鼠神经功能障碍,检测NF-κB阳性表达差异。结果HL组大鼠血清TC、TG、LDL-C水平均显著高于普通组,HDL-C水平显著低于普通组,差异均有统计学意义(P<0.05)。在术后12~72 h,低剂量普罗布考组、高剂量普罗布考组的神经功能评分显著高于缺血再灌注组,高剂量普罗布考组在术后12~72 h的神经功能评分显著高于低剂量普罗布考组,差异均有统计学意义(P<0.05)。低剂量普罗布考组、高剂量普罗布考组脑梗死体积比显著低于缺血再灌注组,且高剂量普罗布考组脑梗死体积比显著低于低剂量普罗布考组,差异均有统计学意义(P<0.05)。低剂量普罗布考组、高剂量普罗布考组在术后12~72 h的脑缺血区NF-κB阳性细胞数量显著低于缺血再灌注组,且高剂量普罗布考组各时间点脑缺血区NF-κB阳性细胞数量均显著低于低剂量普罗布考组,�Objective To investigate the effects of probucol on neurological function,cerebral infarction ratio and nuclear factor-κB(NF-κB)expression after cerebral ischemia-reperfusion in hyperlipidemia(HL)rats.Methods A total of 120 healthy male SD rats were selected and fed for 1 day.They were divided into a normal group(20 rats)and a high-fat group(100 rats).They were fed with normal diet and high-fat die.The HL model was established,and were divided into sham operation group,ischemia-reperfusion group,low-dose probucol group,high-dose probucol group,25 rats in each group.The low-dose probucol group and the high-dose probucol group were given 100 and 500 mg/kg gavage respectively.The sham operation group and the ischemia-reperfusion group were given the same dose of single 0.5%sodium carboxymethyl cellulose by gavage,daily 1 time for 2 consecutive weeks.In addition to the sham operation group,the middle cerebral artery embolism(MCAO)model of rats was prepared in the ischemia-reperfusion group,low-dose probucol group,and high-dose probucol group.The rats were randomly divided into five subgroups(5 rats in each group)according to the different reperfusion time points(6 h,12 h,24 h,48 h,72 h)after cerebral ischemia for 2 h.The cerebral infarction volume ratio was calculated by TTC staining after reperfusion for 24 h after cerebral ischemia for 2 h.The levels of total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C)and high-density lipoprotein cholesterol(HDL-C)in serum were detected,and the neurological dysfunction of rats in each group was evaluated,and the difference in NF-κB positive expression was detected.Results The levels of serum TC,TG and LDL-C in the HL group were significantly higher than those in the normal group,and the levels of HDL-C were significantly lower than those in the normal group,and the differences were statistically significant(P<0.05).The neurological function scores of the low-dose probucol group and the high-dose probucol group at 12-72 h after surgery were signifi
