摘 要:目的探究色氨酸羟化酶2(TPH2)基因多态性与原发性早泄(PPE)的关系。方法选取2017年5月至2019年5月丹东市中心医院诊治的168例PPE患者作为观察组。另选取170例健康体检者作为对照组。采集受试者清晨空腹外周血并提取血清,限制性片段长度多态性-聚合酶链反应(RFLP-PCR)检测TPH2基因rs1007023基因型;调查观察组阴道内射精潜伏期(IELT),检测血清游离睾酮(FT)、黄体生成素(LH)、催乳素(PRL)水平。结果两组TPH2基因的基因型及等位基因分布比较,差异具有统计学意义(P<0.05);与对照组比较,观察组TPH2基因G等位基因频率明显升高,T等位基因频率明显降低,差异具有统计学意义(P<0.05);与对照组比较,观察组IELT明显降低,血清FT、LH、PRL水平明显升高,差异具有统计学意义(P<0.05);与GG基因型比较,TG、TT基因型PPE患者IELT均明显升高,血清FT、LH、PRL水平明显降低,差异具有统计学意义(P<0.05);与TG基因型比较,TT基因型PPE患者IELT明显升高,血清FT、LH、PRL水平明显降低,差异具有统计学意义(P<0.05)。结论PPE患者携带rs1007023 G等位基因的频率显著升高,其不同TPH2基因型间IELT及血清FT、LH、PRL水平有显著差异。Objective To investigate the relationship between tryptophan hydroxylase 2(TPH2)gene polymorphism and primary premature ejaculation(PPE).Methods 168 PPE patients treated in Dandong Central Hospital from May 2017 to May 2019 were selected as the observation group.Another 170 healthy subjects were selected as control group.Early morning fasting peripheral blood was collected and serum was extracted.Restriction fragment length polymorphisms-PCR(RFLP-PCR)were used to detect rs1007023 genotype of TPH2 gene.The intravaginal ejaculation latency time(IELT)in the observation group was investigated,and the serum levels of free testosterone(FT),luteinizing hormone(LH)and prolactin(PRL)were detected.Results The genotypes and alleles distribution of TPH2 gene between the two groups were statistically significant(P<0.05).Compared with the control group,the frequency of G allele of TPH2 gene in the observation group was significantly increased,and the frequency of T allele was significantly decreased,with statistical significance(P<0.05).Compared with the control group,IELT in observation group was significantly decreased,while serum FT,LH and PRL levels were significantly increased,with statistical significance(P<0.05).Compared with GG genotype,IELT was significantly increased in TG and TT genotype,while serum FT,LH and PRL levels were significantly decreased,with statistical significance(P<0.05).Compared with TG genotype,IELT of PPE patients with TT genotype was significantly increased,while serum FT,LH and PRL levels were significantly decreased,with statistical significance(P<0.05).Conclusions The frequency of carrying rs1007023 G alleles in PPE patients increased significantly,and there were significant differences in IELT and serum FT,LH,PRL levels among different TPH2 genotypes.
