摘 要:随着生物制药技术的迅速发展,生物制品的制备规模和质量要求的提高对下游分离纯化工艺提出了新的要求。如何通过分离纯化手段快速获得目标产物并进一步降低成本是现代生物分离技术研究的热点问题。混合模式色谱技术是一种高效的分离纯化方法,一般兼有多种作用模式,并且其蛋白结合量高、选择性好、洗脱条件温和、具有耐盐吸附等多种特性,可减少对料液的预处理操作,从而提高分离效率,降低生产成本。因此,该技术被广泛应用于蛋白质的分离纯化及相关领域中。本文对混合模式色谱技术的发展历程、分离机理、基质、配基类型及其在蛋白分离纯化中的应用等方面进行阐述,对当前混合模式介质的制备及其应用中存在的问题进行概括,希望能够为该项技术在生物制品制备生产中的应用提供一些参考。With the rapid development of biopharmaceutical technology,the improvement of the scale and quality standards of biological prod-ucts requires higher efficiency of downstream separation and purification process.How to obtain efficiently target products and further reduce the cost through separation and purification is a heated issue in biological separation technology.Mixed-mode chromatography is an efficient separation and purification method with multi-mode interactions.This technique has been widely used in the purification of proteins and related fields due to its ad-vantages,including high protein binding capacity,good selectivity,mild elution conditions,salt adsorption resistance and other characteristics.Mean-while,high-efficiency separation and low cost can be obtained in this technology because it can reduce the pretreatment operation of the feed liquid.This paper focused on the development history,separation mechanism,substrate,ligand types of mixed-mode chromatography and its application in protein separation and purification.Furthermore,the present problems in the preparation of mixed-mode media and its application are summarized,it is expected to provide some information on the preparation and production of biological products using mixed-mode chromatography.
