摘  要:目的探讨补肾益肺消癥方抑制特发性肺纤维化病理进程的作用机制。方法以转化生长因子β1(Transforming growth factor-β1,TGF-β1)诱导的A549细胞为模型,以TGF-β1抑制剂为阳性对照,筛选补肾益肺消癥方两种浓度冻干粉干预细胞模型,免疫荧光染色激光共聚焦显微镜下观察SP-C、α平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)及E-钙黏蛋白(E-Cadherin)表达,western-blot检测c-JUN氨基末端激酶(JNK)信号通路相关蛋白表达量。结果模型组较正常组细胞中SP-C、α-SMA表达明显增多(P<0.01),E-Cadherin表达明显降低(P<0.01),模型组细胞中JNK信号通路相关蛋白表达较正常组均明显升高(P<0.05,P<0.01);对照组及中药组较模型组细胞中SP-C、α-SMA表达明显降低(P<0.05,P<0.01),E-Cadherin表达明显升高(P<0.01);对照组及中药组细胞中JNK信号通路相关蛋白表达较模型组均明显降低(P<0.05,P<0.01)。结论补肾益肺消癥方可抑制肺泡上皮细胞的间质转化,并通过减轻持续的内质网应激,抑制Ⅱ型肺泡上皮细胞的凋亡,达到延缓特发性肺纤维化病理进程的治疗作用。Objective To explore the mechanism of Bushen Yifei Xiaozheng Decoction in inhibiting the pathological process of idiopathic pulmonary fibrosis.Methods A549 cells induced by transforming growth factor-β1(TGF-β1)were used as model, and TGF-β1 inhibitor was used as positive control to screen two concentrations of lyophilized powder of Bushen Yifei Xiaozheng to intervene cells.Model, the expression of SP-C,α-smooth muscle actin(α-SMA)and E-Cadherin(E-Cadherin)were observed under the immunofluorescence staining laser confocal microscope, and c-Cadherin was detected by western-blot.Western-blot was used to detect the protein expression of JNK signaling pathway-related.Results Compared with the normal group, the expression of SP-C and α-SMA in the model group was significantly increased(P<0.01),and the expression of E-Cadherin was significantly decreased(P<0.01).The expression of JNK signaling pathway-related proteins in the cells of the model group was higher than that of the normal group.Compared with the model group, the expressions of SP-C and α-SMA in the control group and the traditional Chinese medicinegroup were significantly decreased(P<0.05,P<0.01),and the expression of E-Cadherin was significantly increased.The expression of JNK signaling pathway-related proteins in the cells of the control group and the traditional Chinese medicine group was significantly lower than that of the model group(P<0.05,P<0.01).Conclusion Bushen Yifei Xiaozheng Decoction can inhibit the interstitial transition of alveolar epithelial cells, and by reducing the persistent endoplasmic reticulum stress, inhibit the apoptosis of type Ⅱ alveolar epithelial cells, and delay the pathological process of idiopathic pulmonary fibrosis.Therapeutic effect.
