摘 要:目的通过观察麻黄、桂枝贴敷大椎穴对干酵母诱导大鼠发热模型的退热效应,探讨穴位贴敷退热的作用机制。方法将40只SPF级SD雄鼠随机分为空白组对照组、模型对照组、穴位贴敷组、非穴位贴敷组、阳性对照组,每组8只。对各组大鼠进行预处理,预处理结束后除空白组外其他组利用干酵母制备发热模型,分别测量各组大鼠造模后1~8 h的体温,并于实验结束后,观察各组大鼠肺组织HE染色病理变化,测量各组大鼠肺组织白细胞介素-6(Interleukin 6,IL-6)、白细胞介素-1β(Interleukin-1 receptor, IL-1β)、肿瘤坏死因子(Tumor necrosis factor-α,TNF-α)表达变化。结果造模后发现模型组体温变化明显大于空白组,说明造模成功,干酵母模型发热稳定;体温变化方面,穴位贴敷组、非穴位贴敷组、阳性对照组体温升高均明显小于模型组,差异有统计学意义(P<0.05);组织HE染色病理变化方面,与模型组比较,其余各组大鼠下丘脑和肺组织形态学均有明显的改变;肺组织细胞因子表达变化方面,与模型组比较,穴位贴敷组大鼠肺组织IL-6、IL-1β水平较低(P<0.05),TNF-α水平明显降低(P<0.01);非穴位贴敷组TNF-α水平降低(P<0.05),IL-6、IL-1β水平降低不明显;阳性对照组TNF-α、IL-6水平降低(P<0.05),IL-1β降低不明显。结论麻黄、桂枝贴敷大椎穴可以通过降低肺组织致热细胞因子IL-6、IL-1β、TNF-α的表达,进一步改变肺组织形态,进而达到稳定退热的作用。Objective To observe the antipyretic effect of Mahuang and Guizhion Dazhui acupoint in rats with dry yeast-induced fever model, and to explore the mechanism of antipyretic action of acupoint application.Methods Forty SPF SD male rats were included in the study and randomly divided into blank group, model group, acupoint application group, non-acupoint application group, and tirolin positive control group.Each group was pretreated.After the pretreatment, dry yeast was used to prepare a fever model for the other groups except the blank group, and the body temperature of rats in each group was measured 8 hours after modeling.At the end of the experiment, the pathological changes of HE staining in lung tissues of rats in each group were observed, and the expression changes of IL-6,IL-1β and TNF-α in lung tissues of rats in each group were measured.Results After modeling, it was found that the temperature change of the model group was significantly greater than that of the blank group, indicating that the modeling was successful, and the dry yeast model had a stable fever;in terms of body temperature changes, the increase in body temperature in the acupoint application group, non-acupoint application group, and positive control group was significantly smaller than that in the positive control group.The difference was statistically significant in the model group(P<0.05);in terms of the pathological changes of tissue HE staining, it was also found that compared with the model group, the morphology of the hypothalamus andlung tissue of the other groups of rats had obvious changes;lung tissue cytokines In terms of expression changes, compared with the model group, the levels of IL-6 and IL-1β in the lung tissue of the acupoint sticking group were lower(P<0.05),and the level of TNF-α was significantly decreased(P<0.01);the non-point sticking group The level of TNF-α decreased(P<0.05),but the levels of IL-6 and IL-1β were not significantly decreased;the levels of TNF-α and IL-6 in the positive control group were decr
