摘 要:目的探讨五季(春、夏、长夏、秋、冬)胃肠道(胃、空肠、回肠、结肠)褪黑素(Melatonin,MT)分泌的季节性变化与"脾应长夏"生理机制的关系。方法将60只健康SD大鼠随机分为10组,对照组和模型组各5组,按照春、夏、长夏、秋、冬五个季节分批饲养,每个季节均安排两组,对照组和模型组各6只,模型组行松果腺摘除手术,对照组不做任何处理。分别于五季代表时间点(春分、夏至、立秋前1 d、秋分、冬至)取胃、空肠、回肠、结肠,采用酶联免疫吸附测定法(Enzyme-Linked ImmunoSorbent Assay,ELISA)测定相应组织的MT含量并进行比较。结果(1)在正常生理情况下,空肠和回肠的MT水平均表现为长夏季节明显高于其他季节,差异有统计学意义(P<0.05,P<0.01);胃MT水平五季的季节性变化不明显;结肠MT水平表现为冬季最高。(2)松果腺摘除后,胃、空肠、回肠、结肠组织的MT季节性变化规律均发生了改变。结论胃肠道MT的分泌呈现出明显的季节性波动,长夏季节空肠、回肠MT分泌增高可能是"脾应长夏"的重要生理机制。松果腺参与了胃肠道MT季节性变化规律的调节。Objective To explore the relationship between the seasonal changes of melatonin(MT)secretion in various segments of the gastrointestinal tract(stomach,jejunum,ileum,colon)in the five seasons(spring,summer,late summer,autumn and winter)and the physiological mechanism of“spleen responds to late summer”.Methods 60 healthy SD rats were randomly divided into 10 groups,the control group and the model group with 5 groups each.They were reared in batches according to the five seasons of spring,summer,long summer,autumn and winter.Two groups were arranged in each season.There were 6 rats in the control group and the model group,the model group underwent pineal gland removal surgery,and the control group did not receive any treatment.Stomach,jejunum,ileum,and colon were collected at the representative time points of the five seasons(spring equinox,summer solstice,1 day before the beginning of autumn,autumnal equinox,and winter solstice).Enzyme-linked immunosorbent assay(Enzyme-Linked ImmunoSorbent Assay,ELISA)was used to measure the MT content of corresponding tissues.Results(1)Under normal physiological conditions,MT levels in jejunum and ileum were significantly higher in late summer than other seasons(P<0.05,P<0.01);MT levels in stomach did not change significantly in five seasons;MT levels in colon were the highest in winter.(2)After pinealectomy,the seasonal changes of MT in stomach,jejunum,ileum and colon were changed.Conclusion The secretion of MT in the gastrointestinal tract exhibits obvious seasonal fluctuations,and the increased secretion of MT in the jejunum and ileum during the long summer season may be an important physiological mechanism of“the spleen should be in the late summer”.The pineal gland is involved in the regulation of seasonal changes in MT in the gastrointestinal tract.
