摘  要:目的探讨血管紧张素1-7(Angiotensin1-7,Ang1-7)对盐敏感性高血压大鼠心脏钠泵α1亚单位(Na^(+)-K^(+)-ATPaseα1)的影响。方法选取35只新生雄性Wistar大鼠建立感觉神经损伤性盐敏感高血压大鼠模型,后随机分成7组(n=5):正常对照组、假手术组、模型组、替米沙坦组、雷米普利组、Ang1-7组、A-779组。经哺乳期(3周)后开始测量各组大鼠鼠尾血压,每周1次,8周后剪取大鼠左心室心尖部组织。通过酶联免疫吸附法检测心室肌组织中Ang II和Ang1-7的含量,通过RT-PCR法检测心室肌组织中Na^(+)-K^(+)-ATPaseα1 mRNA的表达,免疫组化法检测Na^(+)-K^(+)-ATPaseα1蛋白的表达。结果第7周时,与正常对照组比较,高盐饲料饲养的各组大鼠鼠尾收缩压均升高(P<0.05),且各高盐饲养组之间无统计学差异(P>0.05),说明造模成功;药物干预2周(第9周)后,与模型组比较,替米沙坦组、雷米普利组及Ang-(1-7)组血压下降(P<0.05),假手术组及A-779组血压无明显差异(P>0.05)。与模型组比较,正常对照组、Ang1-7组、A-779组、替米沙坦组及雷米普利组心室肌组织中Ang1-7浓度增加(P<0.05);与Ang1-7组比较,假手术组及A-779组Ang1-7水平降低(P<0.05)。与模型组比较,正常对照组、替米沙坦组、雷米普利组及Ang1-7组大鼠心室肌组织中Na^(+)-K^(+)-ATPaseα1亚单位mRNA及蛋白表达量降低(P<0.05),A-779组及假手术组无明显差异(P>0.05)。与Ang1-7组比较,A-779组及假手术组表达增高(P<0.05),其它各组无明显差异(P>0.05)。结论盐敏感性高血压大鼠在早期高血压时期心肌钠泵表达会代偿性增加,而Ang1-7对SSH大鼠心室肌Na^(+)-K^(+)-ATP酶α1亚单位mRNA及蛋白的表达有抑制作用。Objective To investigate angiotensin 1-7(Ang1-7)on sodium pumpα1 subunit(Na^(+)-K^(+)-ATPaseα1)in the heart of salt-sensitive hypertensive rats.Methods Thirty-five newborn male Wistar rats were given capsaicinused(50 mg/kg,sc)to produce sensory nerve injury,followed by high-salt feeding to establish hypertension rat model.Rats were divided into normal control,sham-operated,model,temisartan,ramipril,Ang1-7,and A-779 groups(n=5).Rat tail blood pressure was measured once a week after lactation(3 week).The left ventricular tissues were collected at the end of experiment.The levels of Ang Ⅱ and Ang1-7 in ventricular tissues were measured by enzyme-linked immunosorbent assay,and the expression of Na^(+)-K^(+)-ATPaseα1 mRNA and Na^(+)-K^(+)-ATPaseα1 protein were detected by RT-PCR and immunohistochemistry.Results At week 7,compared with the normal control group,the systolic blood pressure of rats in all hypertension rats was increased(P<0.05),indicating successful modeling.Two weeks after pharmacological intervention(week 9),blood pressure decreased in the temisartan,Ramipril,and Ang1-7groups(P<0.05),and There was no difference in blood pressure in the shamoperated and A-779 groups(P>0.05).Compared with the model group,Ang1-7 concentrations in ventricular myocardial tissue were higher in the normal control,Ang1-7,A-779,temisartan,and ramipril groups(P<0.05).Ang1-7 was lower in sham-operated and A-779 groups than Ang1-7 group(P<0.05).Compared with the model group,Na^(+)-K^(+)-ATPaseα1 subunit mRNA and protein in ventricular tissues were decreased in the normal control,temisartan,ramipril and Ang1-7 groups(P<0.05).Compared with the Ang1-7 group,the expression was increased in the A-779 group and the sham-operated group(P<0.05).Conclusion In the salt-sensitive hypertensive rat model,the expression of myocardial sodium pump increased compensatively in the early stage of hypertension,while Ang1-7 may have an inhibitory effect on myocardial sodium pump.
