摘 要:目的探讨巨噬细胞的活化在增生性瘢痕自然演变过程中的作用。方法分别收集临床增生期增生性瘢痕组织(A组,n=15)和减退期增生性瘢痕组织(B组,n=16),用抗体阵列印记膜法检测其炎症相关因子bFGF、IL-12、IL-10、IL-9、IL-1β、TGF-β、IL-6和VEGF的表达,同时用ELISA检测巨噬细胞活化特异性因子IL-10和IL-12的表达。结果抗体阵列检测结果显示,与B组相比,A组中IL-12、IL-6、TGF-β的表达量均明显升高(P<0.05),而IL-10的表达量则明显降低(P<0.05)。ELISA检测结果显示,与B组相比,A组中IL-12的表达量明显升高(P<0.05),而IL-10的表达量则明显降低(P<0.05),并且IL-10表达量在IL-10和IL-12总表达量中所占的比值也明显降低(P<0.05)。结论增生性瘢痕自然演变过程与巨噬细胞向M2型活化过程中的炎症因子表达变化表现出高度的一致性,巨噬细胞的活化程度可以作为确定增生性瘢痕具体临床时期的分子标志之一。Objective To investigate the role of macrophage activation in the natural evolution of hypertrophic scar.Methods The clinical hypertrophic scar tissue at proliferative stage(group A,n=15)and hypertrophic scar tissue at regressive stage(group B,n=16)were collected respectively.The expression of the inflammatory related factors bFGF,IL-12,IL-10,IL-9,IL-1β,TGF-β,IL-6 and VEGF were detected by antibody array imprinting membrane method,and the expression of macrophage activation specific factors IL-10 and IL-12 were detected by ELISA.Results The results of antibody array detection showed that,compared with group B,the expression of IL-12,IL-6 and TGF-βin group A increased significantly(P<0.05),while the expression of IL-10 was significantly decreased(P<0.05).The results of ELISA detection showed that,compared with group B,the expression of IL-12 in group A was significantly higher(P<0.05),while the expression of IL-10 was significantly decreased(P<0.05),and the ratio of IL-10 expression to the total expression of IL-10 and IL-12 was also significantly lower(P<0.05).Conclusion The natural evolution of hypertrophic scar is highly consistent with the expression of inflammatory factors in the process of macrophage activation to M2 type.The activation degree of macrophages can be used as one of the molecular markers to determine the specific clinical stage of hypertrophic scar.
