摘 要:为了解细胞因子IL-4和IL-5在戊型肝炎病毒(HEV)感染中的表达及其作用,以新西兰大白兔为模型,腹腔注射感染HEV-4,攻毒后第7、14、21、28、35天对兔的心、肝、脑、肾等进行组织病理学观察,应用实时荧光定量PCR对肝脏、脑等器官中IL-4和IL-5的表达水平及变化进行检测。结果显示,肝脏中IL-4和IL-5攻毒后第7天mRNA转录水平明显高于对照组,从第14天开始转录水平降低,逐渐降低直到第35天恢复至与对照组一致;脑中IL-4的转录水平在感染初期就明显降低,第28天达到最低值,仅为对照组的10%左右,第35天转录水平恢复至正常。IL-5在感染初期下调,第14天上调,在第35天恢复至正常。表明IL-4和IL-5参与了HEV感染早期IgG抗体产生,肝脏、脑炎症状发生等过程。研究结果为揭示猪戊型肝炎病毒的致病机理以及抗病毒生物制剂研发等提供参考。In order to understand the expression and role of cytokines IL-4 and IL5 in HEV infection,New Zealand white rabbits were selected as a model,and HEV-4 was infected by intraperitoneal injection,on the 7th,14th,21th,28th,and 35th after the challenge.To observe the histopathology of heart,liver,brain,kidney and other tissues,real-time fluorescent quantitative PCR is used to detect the expression levels and changes of IL-4 and IL-5 in liver,brain and other tissues and organs.The results showed that the mRNA transcription levels of IL-4 and IL-5 in the liver were significantly higher than those in the control group on the 7th day,and the transcription levels decreased from the 14th day,and gradually decreased until the 35th day recovered to be consistent with the control group;The transcription level of IL-4 in the brain decreased significantly at the beginning of the infection,reaching the lowest value on the 28th day,only about 10%of the control group,and the transcription level returned to the normal level on the 35th day.IL-5 was down-regulated at the beginning of infection,up-regulated on the 14th day,and returned to normal levels on the 35th day.It showed that IL-4 and IL-5 are involved in the process of IgG antibody production in the early stage of HEV infection,liver and brain inflammation.This research laid the foundation for revealing the pathogenic mechanism of porcine hepatitis E virus and the development of antiviral biological agents.
