摘 要:采集广西25个猪场的病料,对猪流行性腹泻病毒(PEDV)阳性组织样品进行全S基因扩增。将41株全S基因进行序列比对及遗传进化分析,41株PEDV的S基因之间核苷酸同源性为94.8%~100%,与参考毒株核苷酸同源性为89.3%~99.4%。S基因进化树图谱显示,广西当前流行的PEDV可分为2个谱系。Attenuated-DR13、CV777和该研究中的CHGXLC055-92013、CHGXLC055-52013、CHGXGP035-72013和CHGXGP035-82013为Cluster1;Cluster3包括该研究另外37株毒株、韩国株Spk1、HuN和日本株NK等参考毒株。表明广西大部分毒株S基因与欧洲株CV777、LZC等国内早期毒株存在较大的差异,进化树亲缘关系比较远;在抗原表位区域存在较大变异,当前的CV777疫苗株在防控PEDV入侵时可能不会产生良好的免疫保护效果。To provide a scientific basis for better prevention and control of PEDV and its current characteristics of genetic evolution in Guangxi,41 PEDV field strains were isolated from 25 farms in Guangxi.S gene was amplified from the 41 PEDV field strains,cloned,sequenced and compared with the other reference strains in GenBank.The S gene of 41 PEDV strains had 94.8%-100%nucleotide identity among them and 89.3%-99.4%with the domestic and foreign reference strains.Phylogenetic analysis of S gene indicatied that Guangxi strains could be divided into two groups.Cluster 1 consisted of four strains from our study(CHGXLC055-92013,CHGXLC055-52013,CHGXGP035-72013 and CHGXGP035-82013)and Attenuated-DR13,European strains CV777.Other 37 strains and Korean strains(Spk1),HuN,Japan strains(NK)were in a group 3.It has a great difference between strains in Guangxi and European strains(CV777)and early domestic strains(LZC).By the epitope region comparing with Chinese vaccine strain CV777,we conclude that the current vaccine has not played a effective role in immunization.
