祛风通络方对高糖诱导人肾小球系膜细胞炎症反应的影响

摘　　要：目的观察祛风通络方对高糖诱导人肾小球系膜细胞炎症反应的影响。方法体外培养人肾小球系膜细胞株,将其分为正常组(低糖DMEM培养液+正常大鼠血清)、模型组(高糖DMEM培养液+正常大鼠血清)、抑制剂组(高糖DMEM培养液+正常大鼠血清+5μmol/L SB203580预处理)、祛风通络组(高糖DMEM培养液+服用祛风通络方的大鼠血清)。采用酶联免疫吸附试验检测白细胞介素(IL)-1β、IL-6、肿瘤坏死因子-α(TNF-α)、转化生长因子β1(TGF-β1)的水平,Western blot检测p38丝裂原活化蛋白激酶(p38 MAPK)、细胞间黏附分子-1(ICAM-1)、单核细胞趋化蛋白-1(MCP-1)的表达水平。采用RT-PCR测定IL-1β、IL-6、TNF-α、TGF-β1、p38 MAPK、ICAM-1、MCP-1的mRNA表达情况。结果模型组IL-1β、IL-6、TNF-α、TGF-β1、p38 MAPK、ICAM-1、MCP-1、水平及其mRNA表达均高于正常组,差异有统计学意义(P<0.05)。祛风通络组IL-1β、IL-6、TNF-α、TGF-β1、p38 MAPK、ICAM-1、MCP-1水平及其mRNA表达均低于模型组,差异有统计学意义(P<0.05)。结论祛风通络方可能通过抑制p38 MAPK信号通路降低高糖诱导人肾小球系膜细胞炎症因子的表达,从而减轻炎症反应。Objective To investigate the effects of Qufeng Tongluo Fomula on the inflammatory responce of human mesangial cells induced by high glucose.Methods In vitro culture of human mesangial cells,they were divided into normal group(low-glucose DMEM culture medium+normal rat serum),model group(high-glucose DMEM culture medium+normal rat serum),inhibitor group(high-glucose DMEM culture medium+normal rat serum+5μmol/L SB203580 pretreatment),Qufeng Tongluo group(high-glucose DMEM culture medium+serum of rat taking Qufeng Tongluo Fomula).The levels of interleukin(IL)-1β,IL-6,tumor necrosis factor-α(TNF-α),and transforming growth factor-β1(TGF-β1)were detected by enzyme-linked immunosorbent assay.Western blot was used to detect the expression levels of p38 mitogen activation protein kinase(p38 MAPK),intercelluar adhesion molecule-1(ICAM-1),and monocyte chemotactic protein-1(MCP-1).The mRNA expressions of IL-1β,IL-6,TNF-α,TGF-β1,ICAM-1,MCP-1,and p38 MAPK were determined by RT-PCR.Results The levels and mRNA expression of IL-1β,IL-6,TNF-α,TGF-β1,ICAM-1,MCP-1,and p38 MAPK in model group were higher than those in normal group,the differences were statistically significant(P<0.05).The levels of IL-1β,IL-6,TNF-α,TGF-β1,ICAM-1,MCP-1,and p38 MAPK and their mRNA expression in Qufeng Tongluo group were lower than those in model group,and the differences were statistically significant(P<0.05).Conclusion Qufeng Tongluo Formula may reduce the expression of inflammatory factors in human glomerular mesangial cells induced by high glucose by inhibiting the p38 MAPK signaling pathway,thus alleviating the inflammatory response.
