摘  要:旨在通过测定注射黄芪多糖(Astragalus polysaccharides, APS)后热应激奶牛血清代谢组学,分析血清差异代谢物及代谢通路,为APS缓解奶牛热应激作用提供参考依据。选取6头健康的荷斯坦奶牛,颈部肌肉注射APS注射液(30 mL/d),连续注射4 d,停止3 d为1个周期,试验期共3个周期。试验期前后采集奶牛血清样本,利用气相色谱和质谱(GC-MS)联用代谢组学技术分析奶牛血清代谢质谱,并进行主成分分析(PCA)和正交偏最小二乘方-判别分析(OPLS-DA)。结果表明,共鉴定出27种差异代谢物,9种内源性代谢物可作为APS缓解奶牛热应激的潜在标志物,其中2-酮丁酸、甘氨酸、乳酸、赖氨酸、苯丙氨酸、色氨酸、酪氨酸上调,3-氨基异丁酸、果糖显示下调。此外,APS通过氨基酸代谢、嘌呤代谢、谷胱甘肽代谢等途径影响热应激奶牛的代谢平衡。本试验结果为研究APS缓解奶牛热应激的生理机制提供参考。Serum metabonomics and metabolic pathways of heat stressed dairy cows after injection of Astragalus polysaccharides(APS) were analyzed to provide reference for APS on heat stress cows.Six healthy dairy cows were injected with APS injection(30 mL/d) intramuscularly.During the experiment, APS was injected continuously for 4 d and stopped for 3 d as a cycle.Before and after the experimental period, serum samples were collected and analyzed by gas chromatography-mass spectrometry(GC-MS) combined with metabonomics.Principal component analysis(PCA) and orthogonal partial least squares discriminant analysis(OPLS-DA) were performed.The results showed that 27 different metabolites were identified, nine endogenous metabolites could be used as potential markers for APS to alleviate heat stress in dairy cows.Among them, 2-ketobutyric acid, glycine, lactic acid, lysine, phenylalanine, tryptophan and tyrosine were up-regulated, while 3-aminoisobutyric acid and fructose were down-regulated.Moreover, APS could affect the metabolic balance of heat stressed cows through amino acid metabolism, purine metabolism and glutathione metabolism.The results of this study provide a reference for the physiological mechanism of APS to alleviate heat stress in dairy cows.
