摘 要:采用10,50,100 mg/L甘草查尔酮A处理蹄真皮炎性细胞24 h后,测定细胞上清液中TNF-ɑ、IL-1β、IL-6水平及SOD、MDA含量,Western blot法检测ERK、JNK、p38、IκBα及p65蛋白表达水平。结果显示,甘草查尔酮A可显著降低TNF-ɑ、IL-1β、IL-6水平,提高SOD活性并降低MDA含量,抑制ERK、JNK、p38、IκBα及p65蛋白磷酸化程度。这表明甘草查尔酮A对于LPS诱导的奶牛蹄真皮细胞炎性反应具有一定的抑制作用,且其抗炎作用可能与MAPK及NF-κB信号通路有关。After treated with 10,50,100 mg/L licorice chalcone A on inflammatory hoof dermal cells for 24 h, the levels of TNF-ɑ,IL-1β,IL-6,SOD and MDA in the cell supernatant was detected, the protein levels of ERK,JNK,p38,IκBα and p65 were detected by Western blot.The results showed that licorice chalcone A significantly reduced the levels of TNF-ɑ,IL-1β and IL-6,increased SOD activity, reduced MDA content, and inhibited the phosphorylation of ERK,JNK,p38,IκBα and p65.The results indicated that licorice chalcone A had a certain inhibitory effect on LPS-induced inflammatory response of dairy cow hoof dermal cells, and its anti-inflammatory effect might be related to MAPK and NF-κB signaling pathways.
