摘  要:目的:观察艾灸对APP/PS1双转基因小鼠海马及前额叶皮质线粒体嵴结构蛋白MIC10、MIC60的改善作用,探讨艾灸保护线粒体嵴结构,改善阿尔茨海默病(AD)等神经退行性病变小鼠脑区线粒体能量代谢的相关机制。方法:将6月龄APP/PS1小鼠随机分为模型组、艾灸组,同月龄野生型C57BL/6J小鼠设为正常对照组。艾灸组施以百会、涌泉艾灸治疗。治疗结束后,采用Morris水迷宫定位航行实验测试各组小鼠空间学习记忆能力的变化,评价艾灸的治疗效应;采用免疫荧光染色法,采用CD46抗体标记MIC10,Mitofilin抗体标记MIC60,观察海马各区(DG、CA1、CA3)及前额叶皮层线粒体嵴结构蛋白MIC10、MIC60表达的变化。结果:与正常对照组比较,模型组小鼠隐藏平台平均逃避潜伏期延长(P<0.01);与模型组比较,艾灸组小鼠平均逃避潜伏期显著缩短(P<0.01)。模型组小鼠海马(DG、CA1、CA3)及前额叶皮质层MIC10、MIC60表达量较正常对照组均显著减少(P<0.01,P<0.05)(前额叶皮质层MIC60除外),艾灸后,各区MIC10、MIC60蛋白表达量均增多(P<0.01,P<0.05)(海马CA1、CA3区MIC60除外)。结论:艾灸对AD学习记忆能力的改善可能与其增加海马及前额叶皮质区线粒体嵴结构蛋白、保护线粒体结构和功能、改善能量代谢有关。Objective:To observe the effects of moxibustion on mitochondrial cristae remodeling protein MIC10,MIC60 in hippocampus and prefrontal cortex of APP/PS1 double-transgenic mice,and to explore the related mechanism of moxibustion in protecting the structure of mitochondrial cristae and improving mitochondrial energy metabolism in brain regions of mice with neurodegenerative diseases such as Alzheimer’s disease(AD).Methods:SPF level APP/PS1 mice aged 6 months were randomly divided into model group and moxibustion group,and SPF level wild-type C57 BL/6 J mice aged the same month were set as normal control group.The moxibustion group was treated with Baihui(GV 20)and Yongquan(KI 1).After treatment,Morris water maze navigation experiment was used to test the spatial learning and memory ability of mice in each group to evaluate the therapeutic effects of moxibustion.CD46 and Mitofilin were used to mark MIC10 and MIC60 respectively in immunofluorescence staining to observe the expressions of mitochondrial cristae remoldling protein MIC10 and MIC60 in the hippocampus(DG,CA1 and CA3)and prefrontal cortex.Results:Compared with the normal control group,the average escape latency of the hidden platform in the model group was prolonged(P<0.01);the mean escape latency of moxibustion group mice was significantly shortened after treatment(P<0.01).The results of immunofluorescence showed that the expressions of MIC10 and MIC60 in the hippocampus(DG,CA1,CA3)and prefrontal cortex of mice in the model group were lower than that of the normal control group(P<0.01,P<0.05)(except prefrontal cortex MIC60).After treatments,the protein levels of MIC10 and MIC60 was increased(P<0.01,P<0.05)(except hippocampus CA1,CA3 MIC60).Conclusion:The improvement of AD learning and memory ability by moxibustion may be related to the increase of mitochondrial cristae remodeling proteins in hippocampus and prefrontal cortex,the protection of mitochondrial structure and function,and the improvement of energy metabolism.
