摘 要:目的:观察羌葛真武汤对慢性心力衰竭(CHF)大鼠AVP-AVP受体-AQP调节系统的作用。方法:Wistar大鼠随机分为空白组、模型组、呋塞米组、芪苈强心胶囊组、羌葛真武汤组、真武汤组。除空白组之外,其余动物腹腔注射阿霉素,建立CHF大鼠模型。给药28d后观察一般情况、大鼠心功能(LVEF、LVFS、LVEDV、LVESV);ELISA法检测血浆中NT-proBNP和AVP含量;免疫组化染色评价肾中AQP2改变;Western Blot检测肾脏中AQP2和V2R蛋白水平表达。结果:与模型组比较,羌葛真武汤组显著降低模型大鼠LVEDV和LVESV(P<0.05),升高LVEF、LVFS(P<0.05);降低大鼠血浆中NT-proBNP、AVP表达(P<0.05),降低大鼠肾脏中AQP2表达(P<0.05),降低大鼠肾脏中AQP2蛋白表达(P<0.05)。结论:羌葛真武汤可以降低CHF大鼠的病死率,减轻心脏负荷,其机制可能与调节AVP-AVP受体-AQP调节系统有关,这可能是其改善心功能和水液代谢的重要机制之一。Objective:To observe the effect of Qiangge Zhenwu Decoction on the AVP-AVP receptor-AQP regulatory system in rats with chronic heart failure(CHF).Methods:Wistar rats were randomly divided into blank group,model group,furosemide group,Qiliqiangxin group,Qiangge Zhenwu Decoction and Zhenwu Decoction group.Except for the blank group,the other animals were injected with adriamycin into the abdominal cavity to establish a CHF rat model.After 28 days of administration,the general condition and cardiac function(LVEF,LVFS,LVEDV,LVESV)of the rats were observed.The ELISA method was used to detect plasma NT-proBNP and AVP content,immunohistochemical evaluation of AQP2 changes in the kidney;Western Blot detection of AQP2 and V2 R protein expression in the kidney.Results:Ultrasound showed that compared with the model group,Qiangge Zhenwu Decoction could reduce LVEDV and LVESV in rats with heart failure(P<0.05),increase LVEF and LVFS(P<0.05);reduce the expression of NT-proBNP and AVP in rat plasma(P<0.05)and the expression of AQP2 in rat kidneys,and decrease the expression of AQP2 protein in rat kidneys(P<0.05).Conclusion:Qiangge Zhenwu Decoction can reduce the mortality rate of rats and reduce the heart load.The mechanism may be related to the regulation of the AVPAVP receptor-AQP regulatory system,which may be one of the important mechanisms for improving cardiac function and water metabolism.
